Objective To investigate the effects of Low density lipoprotein receptor-related protein-1(LRP1)gene on prostate cancer.Researchs were performed by validation of ChinaPca GWAS data,immunohistochemistry and public database analysis.Methods LRP1 gene that might be related to PCa was selected based on the results of the whole exome sequencing in 26 guangxi zhuang people with prostate cancer.Nine single nucleotide polymorphisms(SNPS)of LRP1 were verified in the GWAS data of ChinaPCa.Immunohistochemical experiments were performed on 12,18 and 4 cases of PCa tissues、paracancer tissues and prostatitis tissues respectively.LRP1 gene expression data from the cancer and tumor gene atlas(TCGA)database were analyzed.The LRP1 gene expression data from TCGA and GTEx database were drawn into boxplot.Construct a protein-protein interaction network(PPI)diagram on the STRING website andverify the relevant gene associations.SPSS and STATA software were used for statistical analysis.Results ChinaPCa GWAS data validation results show that rs11172113 of LRP1 may be correlated with PCa(P=0.083).And the TC genotype was significantly associated with PCa risk(OR=1.27,95%CL,1.066-1.505,P=0.007).Immunohistochemistry experiment and statistical analysis of TCGA database showed no statistical difference in the expression of LRP1 in PCa and adjacent tissues(P=0.167 and P=0.729,respectively).The drawing of TCGA and GTEx database showed that LRP1 expression was decreased in cancer tissues(P < 0.05).PPI network studies showed that LRP1 was associated with PLAT,PDGFB and PDGFRB,which are related to prostate cancer pathways(P < 0.05).Conclusion The rs11172113 of LRP1 gene is related to prostate cancer,and the low expression of LRP1 gene in prostate cancer tissue may be related to prostate cancer.LRP1 may impact the prostate cancer-related pathways by regulating related genes.
