In this paper,the extraction conditions of Grifola frondosa polysaccharides were optimized by orthogonal test.At the same time,the ethanol precipitation conditions of the extracts were screened.Finally,the optimal conditions for extracting and extracting polysaccharides from Grifola frondosa were determined.The extraction temperature was 100 ?.The time was 1 h,the ratio of material to liquid was 1?10;the extraction was performed 3 times,the final concentration of ethanol precipitation was 85%,and the concentration ratio was 500 mg/mL.The physicochemical properties of the crude polysaccharide of Grifola frondosa were determined,wherein the neutral sugar content was 43.68%.The crude polysaccharide of Grifola frondosa was purified by ion exchange resin,and its physical and chemical properties were determined,wherein the neutral sugar content was 74.91%.It was demonstrated that the purification process can increase the neutral sugar content.The purified Grifola frondosa polysaccharide was separated by molecular weight using ultrafiltration,and the molecular weights were >10 kDa,1-10 kDa and <800 Da,respectively.The physicochemical properties of the three parts were determined,and the constituent sugars were analyzed by pre-column derivatization.The molecular weight distribution was determined by GPC method,and the linkage structure was determined by methylation method.In this paper,the anti-tumor activity and immunological activity of the three parts of Grifola frondosa polysaccharide,purified Grifola frondosa polysaccharide and ultrafiltration were verified.The results showed that the purification process improved the neutral sugar content and improved its activity.A small molecular weight fraction with higher activity is screened.In this paper,the active ash tree flower polysaccharide was modified by FITC and then subjected to iodine substitution reaction.The iodine content was determined to determine the polysaccharide content.The rats were given iodine-active ash tree flower polysaccharide by intragastric rats,and the rats were treated with ocular vein at fixed time.Blood was taken and the organ was taken for 3 h to determine the iodine content by ICP-MS to calculate the bioavailability of Grifola frondosa polysaccharide.
