| Objectives:Gallbladder stones are common and frequently-occurring diseases in the clinic.There are many reasons for the formation of stones,including systemic and local factors.Local infection and inflammation of the gallbladder are one of them.Inflammation interacts with stones.This study intends to construct a canine gallbladder stone model,and explore the model construction method,summarize the experience and lessons for reference for similar experimental research;on this basis,add the flavor of Yinchenhaotang to urinate gallbladder stone inflammation model,investigate gallbladder stones and gallbladder The initial mechanism of interaction between inflammation.Methods:After 19 weeks of adaptive feeding,the Beagle was implanted with stones in the gallbladder and injected with appropriate concentration of dilute hydrochloric acid to cause acute inflammatory reaction.A catheter was implanted into the gallbladder and constructed artificially.The channel was established to establish a model of canine gallstone with acute cholecystitis.After 5 days,biochemical and B-ultrasound were reviewed to test whether the model was successful.The successful modeling dogs were divided into 3 groups by random number method,with 4 in each group.The drug was administered at the 3rd week.The blank model group was given saline irrigation,the oral control group was given the Jiawei Yinchenhao soup,and the gallbladder lavage treatment group was given the flavored Yinchenhao soup.After 10 days of continuous administration,each Groups of canine femoral blood were drawn and the gallbladder tissues were harvested.The levels of white blood cells and liver function were detected.The levels of IL-1β in canine serum,bile and gallbladder mucosa were detected by ELISA.Results:Modeling results:12 dogs were successfully modeled.The other 7 dogs either died or were infected,or the catheter was detached,so the drug-filled test could not be performed.The modeling success rate was 63%.The results of the experiment of flavoring Yinchen Artichoke soup:1 general situation:the dog weight loss,poor spirit,low voice,weak tail,activity,less food intake,stool thinning,etc.After the administration,the model group No intervention was slower;some dogs were aggravated;2 white blood cells:between groups,the model group had the highest number of white blood cells,the treatment group had the lowest number,the treatment group was comparable with the model group(P<0.05);3 serum,bile,gallbladder mucosa IL-1β content:Compared with the model group,the IL-1β content in the treatment group and the control group was significantly lower than that in the model group(P<0.05),which was statistically significant;however,the treatment group was not comparable with the control group(P>0.05).4 Safety:During the experimental study,there were no obvious abnormalities in the general conditions,diet,and two stools in the experimental groups.There was no obvious adverse reaction in the gallbladder and oral administration,and the safety was confirmed.Conclusions:The model of canine gallbladder stone was successfully established.The experiment confirmed that Jiawei Yinchenhao Decoction can reduce the number of serum white blood cells,reduce the content of IL-1β in serum,bile and gallbladder mucosa,improve the general condition of dogs,and have certain curative effect.This method can provide a reference for patients with clinical treatment of gallstones with acute cholecystitis. |
