| Objective: To investigate the clinical expression characteristics and biological function of LncRNA LINC01606 associated with gastric cancer and its potential clinical value in the diagnosis and prognosis of gastric cancer.Methods:(1)According to our previous data of microarray,LncRNA LINC01606 was screened out.The expression of LINC01606 was further verified in gastric cancer and adjacent normal tissues(n=75),gastric cancer cell lines and normal gastric mucosal epithelial cell line by RT-qPCR.(2)Combined with the clinicopathological data of the patients,the relationship between the expression of LINC01606 and clinicopathological features in gastric cancer was analyzed.(3)Thedifference of plasma LINC01606 expression between gastric cancer patients(n=48)and healthy controls(n=20)was detected by RT-qPCR,and the relationships between plasma LINC01606 expression and tumor size and TNM stage were analyzed.ROC curve was used to evaluate the clinical value of plasma LINC01606 in diagnosis of gastric cancer.(4)Online tools were used to predict that LINC01606 may be as ceRNAcombinedwith miR-423-5p,and may be involved in the regulation of Wnt signaling pathway,which was confirmed by in vitro experiments.(5)The level of mRNA,the key molecule of Wnt signaling pathway,was detected by RT-qPCR in gastric cancer and adjacent normal tissues(n=75),and correlation analysis was used to speculate whether there was a mutual regulation relationship between the signaling pathway and LINC01606.(6)In vitro experiment,after specificblocking Wnt/β-catenin signaling pathway by JW74,the expression of LINC01606 and miR-423-5p,the level of Wnt3 a mRNA and protein were detected,and the invasion and migration ability of gastric cancer was evaluated by transwell and scratch assay.Result:(1)The expression level of LINC01606 was increased in gastric cancer tissues and gastric cancer cells.(2)The expression of LINC01606 in gastric cancer was positively correlated with lymph node metastasis and TNM stage,negatively correlated with serum total protein and serum albumin,and was an independent risk factor for metastasis and invasion of gastric cancer.(3)The expression of plasma LINC01606 in patients with gastric cancer was higher than that in healthy controls,and the expression level was related to TNM stage,but not to tumor size.The area under the plasma LINC01606 ROC curve was 0.871,the sensitivity was 89.13%,and the specificity was 75.00%.It had certain clinical value in the diagnosis and prognosis of gastric cancer.(4)The expression level of mRNA,the key molecule of Wnt/β-catenin signaling pathway in gastric cancer,was higher than that in adjacent normal tissues,and was positively correlated with the expression of LINC01606.(5)Online tools predictedthat miR-423-5p had complementary binding sites with LINC01606 and Wnt3 a mRNA.(6)JW74 inhibited Wnt/β-catenin signaling pathway,decreased the expression of LINC01606 and miR-423-5p,decreased the expression of Wnt3 a mRNA and protein,and weakened the ability of invasion and migration of gastric cancer cells.Conclusion:(1)LINC01606 was highly expressed in gastric cancer tissues and gastric cancer cell lines,and was positively correlated with lymphatic metastasis and TNM stage of gastric cancer,and negatively correlated with serum total protein and serum albumin in patients with gastric cancer.The results suggest that LINC01606 may be involved in the regulation of invasion and metastasis of gastric cancer.(2)The level of plasma LINC01606 expression has potential clinical value in the diagnosis and prognosis of gastric cancer.The higher the level of LINC01606 expression was in serum of patients with gastric cancer,the later the tumor stage was,and the worse the prognosis was.(3)LINC01606 may promote the expression of Wnt3 a through ceRNA mechanism combined with miR-423-5p,and enhance Wnt/β-cateninsignal,so that Wnt/β-catenin signaling pathway is overactivated in patients with gastric cancer,thus promoting the invasion and metastasis of gastric cancer.On the contrary,the inhibition of Wnt signaling pathway can also decrease the expression of LINC01606. |
