ObjectiveThis paper aims to explore whether Wenyang Tongmai Fang can protect the myocardial tissue of rats with ischemic-reperfusion via JAK2/STAT3 signal transduction pathway.MethodsThirty healthy male SD rats were divided into 5 groups randomly:sham operation group(sham group),ischemia-reperfusion group(IR group),low-dose group(L group),middle-dose group(M group),high-dose group(H group).After 14 days of intragastric administration,the model of myocardial ischemia-reperfusion injury was established by ligation of the left anterior descending coronary artery for 30 minutes and then releasing the ligated artery for 120 minutes.The serum levels of CK-MB,LDH and AST were detected by automatic biochemical analyzer.The ST-segment reduction of the rats was counted in the electrocardiogram.Pathological changes in cardiac tissue were observed by HE staining.The expression of AQP1 and AQP4 in myocardial tissue was detected by immunohistochemical staining.The protein expression of JAK2,p-JAK2,STAT3,p-STAT3,AQP1 and AQP4 was detected by Western-blot method.ResultsCompared with the sham group,the levels of CK-MB,LDH and AST in the serum of the IR group were significantly increased(P<0.01),and the ST-segments of the electrocardiogram were significantly elevated after ischemia for 30 minutes and reperfusion for 120 minutes(P<0.01).HE staining showed that the myocardial cells in the IR group were severely damaged,with obvious swelling and cytoclasis,and a large number of neutrophil infiltration.The positive expression of AQP1 and AQP4 in immunohistochemistry was significantlyincreased(P<0.01).The results of western-blot showed that the expressions of JAK2,p-JAK2,STAT3,p-STAT3,AQP1 and AQP4 were significantly increased(P<0.01).Compared with the IR group,the serum levels of CK-MB,LDH and AST were significantly decreased(P<0.01),and the ST-segments were decreased after 30 minutes ischemia and 120 minutes reperfusion,and the difference was significant(P<0.01),Pathomorphology showed that myocardial cells' damage was alleviated to varying degrees in HE staining.The positive expression of protein AQP1 and AQP4 in immunohistochemistry was significantly reduced,and the difference was statistically significant.Western-blot results showed that the expression of JAK2,p-JAK2,STAT3 and p-STAT3 was generally increased,especially the expression of p-JAK2 and p-STAT3 was significantly higher than that of IR group except for p-STAT3 of high-dose group(P<0.01).AQP1 and AQP4 expression levels were reduced to varying degrees,and the difference was significant.ConclusionsWenyang Tongmai Fang can protect the myocardium by reducing the level of myocardial enzymes in the serum and the elevation of ST-segment of electrocardiogram of myocardial ischemia-reperfusion rats,and improving the pathological changes of myocardial tissue.The myocardial protection is possibly achieved by activating the JAK2/STAT3 signaling pathway,while associated with the inhibition of the expression of AQP1 and AQP4.
