Objective Based on the former studies of the structure and function changes of moxibustion treated knee osteoarthritis?KOA?model rats,and in order to discuss the different effect of moxa smoke and heat in the moxibustion treatment,in this study,to observe the variation of surface enhanced raman spectrum?SERS?of serum,synovial tissue and local muscular tissues of“housanli”area after treated to KOA model rats with moxibustion,moxa smoke and moxibustion heat seperately.METHODS 60 healthy male SD rats with a body mass of 150±20g were selected as the normal control group?referred to as the"normal group"?.The remaining rats were injected with a mixture of 0.03 mol/L l-cysteine and 4%papain?1:2 ratio?in the articular cavity of the left knee to prepare KOA model rats.40 rats successfully replicated in the model were randomly assigned to the model control group?"model group"?,the moxibustion"housanli"group?"moxibustion group"?,the moxibustion and smoke moxibustion group?"moxibustion group"?and the moxibustion and smoke removal group?"smoke removal group"?,with 10 rats in each group.Rats in the moxibustion group,the moxibustion group and the non-smoking group were given 30d intervention in the"hou sanli"point area,once a day for 30min each time.The normal group and the model group were not given any treatment.The SERS detection was applied to analyze the general situation of Raman spectrum peak in serum of rats,synovial tissue of knee joint and muscle tissue in"post sanli"acupoint.OriginPro 8.5 and Photoshop CC were used for image processing,NGLabSpec,and Graphpad Prism 6 were used for data processing and statistics,and the differences of Raman frequency shift and characteristic peak in each group were compared.Results?1?Comparison of SERS spectrum peaks of serum of rats in each groupThe serum of the normal group and the model group showed significantly different characteristic peaks at 495 cm-1,883 cm-1,1447 cm-1,etc.Meanwhile,the model group produced a peak with strong and specific enhancement at 783 cm-1.Compared with the model group,the relative intensity of SERS spectrum in the serum of rats in the moxibustion group,the moxibustion group and the tobacco removing group was significantly lower than that of the model group.The characteristic peaks not found in the model group appeared in the three groups at 460cm-1 and 735cm-1,and the relative intensity increased after the intervention at 1447cm-1.?2?Comparison of SERS spectrum peaks of muscle tissues in"housanli"point area of rats in each groupThere was a significant difference in Raman strength between the normal group and the model group in terms of homogeneous characteristic peaks of muscle tissues in the"posterior sanli"point area.In particular,the peak strength of several homogeneous characteristic peaks represented by 950 cm-1 and 1237 cm-1 in the model group was significantly increased,and a specific spectral peak was produced at 513 cm-1that was not found in the normal group.The muscle tissues of the model group,the moxibustion group,the moxibustion group and the non-smoking group showed characteristic peak-to-peak positions at 480 cm-1,620 cm-1,790cm-1 and 1213cm-11 in the three groups,and the characteristic peak-to-peak strength of the treatment group was significantly higher than that of the model group,and the peak strength of the moxibustion group was the highest,followed by the non-smoking group,and then the moxibustion group.At the peak position of 1125cm-1,the peak strength of the model group was higher than that of the moxibustion group,the moxibustion group and the smoke removal group.?3?comparison of SERS spectrum peaks of synovial tissues of knee joints of rats in each group Compared with normal group,model group knee joint synovial tissue in the most common characteristic peak of peak intensity change is not obvious,but in a 655 cm-1,950 cm-1,1447cm-1,1335 cm-1 represented several spectral peak homogeneity characteristic peaks show that the peak strength of significant differences,and in 655 cm-1,950 cm-11 for a significant rise in the relative content of model group,and 1447 cm-1 and 1335 cm-11 model is the relative strength of two groups decreased significantly.Compared with model group,moxibustion and moxa smoke and smoke moxibustion after the intervention,three sets of knee joint synovial tissue of rats in 870 cm-1,1060 cm-1,1390 cm-1,1185 cm-1,1770 cm-1,1540 cm-1,1895 cm-1,1800 cm-11 the characteristics of the model group appeared no fengfeng,and significant peak position of the peak strength changes.Conclusion?1?Treated KOA model rats with moxibustion,moxa smoke and moxibustion heat seperately could induce special SERS spectrum expression of the serum,synovial tissue and local muscular tissue of“housanli”area,which indicate that some new special materials occurred.?2?After treated with moxibustion,moxa smoke and moxibustion heat,variation expression of homogeneous characteristic peak in serum,synovial tissue and local muscular tissue of“housanli”area were distinguished,which indicate that during the moxibustion,smoke and heat work together,and combination of smoke and heat were benefit to release effective substance.
