Objective:To investigate the protective effect of Empagliflozin preconditioning on myocardial ischemia/reperfusion injury in isolated rat hearts.Methods:32 adult Wistar rats were randomly divided into control group(K-H buffer solution continuous perfusion 120 min),ischemia-reperfusion group(perfusion with K-H buffer solution for 30 min,ischemia for 30 min,reperfusion for 60 min),EMPA a group(perfused with K-H buffer solution for 10 min,perfused with EMPA 0.1 ?mol/L K-H buffer solution for 20 min,ischemia for 30 min,and reperfusion for 60 min),and EMPA b group(perfused with K-H buffer solution for 10 min,containing EMPA 1 ?mol/L K-H buffer solution perfusion for 20 min,30 min ischemia,60 min reperfusion).A total of 4 groups,8 in each group.The modified Langendorff isolated cardiac perfusion model was established to measure the isolated heart perfusion,ischemia/reperfusion(I/R)and isolated rat myocardium LVDP,+dp before and after ischemia at 10,30 and 60 min./dtmax,-dp/dtmax,HR.Coronary reflux was collected to determine CK,LDH,and MDA contents.Myocardial ischemic area was measured by TTC staining.Results:LVDP,+dp/dtmax,-dp/dtmax,and HR during cardiac perfusion were monitored and recorded in all groups.The results showed that continuous perfusion of perfusate kept the cardiac function within the normal range within a certain time range.After a certain period of time after ischemia/reperfusion,heart function index was significantly lower than that before ischemia;EMPA could partially inhibit I/R injury-induced reduction of cardiac function.In the experiment,cardiac coronary pumping fluid was collected 1 min before ischemia and 60 min after reperfusion,and CK,LDH,and MDA were detected.The results showed that the isolated heart was perfused with continuous flow perfusion,CK,LDH,and MDA.Little change:CK,LDH,and MDA increased significantly after ischemia/reperfusion;EMPA could reduce the release of CK,LDH,and MDA caused by I/R injury.TTC staining was used to monitor the percentage of myocardial infarct size.The results showed that:In the case of continuous perfusion of isolated heart,the myocardial tissue showed few infarcts.After ischemia/reperfusion,myocardial ischemia was evident;EMPA could reduce I/R.The ischemic area of the isolated heart.Compared with the above data,the effect of 1 ?mol/L EMPA was more obvious than 0.1 ?mol/L EMPA.Conclusions:EMPA can inhibit the decrease of cardiac function index induced by I/R injury,reduce the area of myocardial infarction and restore the systolic and diastolic function of ischemic myocardium,and EMPA has a certain myocardial protective effect.
