Objective:To investigate the relationship between GSTM1,GSTT1,CAT gene polymorphism and the risk of abnormal liver function in a northern Han Chinese poplationt reated with antiepileptic drugs.Methods:According to the level of liver function indicators,a total of 153 epileptic patients diagnosed in neurology Department,No1 Hospital of Jilin University were divided into normal liver function group(control group)of 57 patients and abnormal liver function group(experimental group)of 96 patients.All the patients have already performed EEG,cranial MRI or CT scan,liver function and other related tests.At the meanwhile,the general information such as age,gender,height,weight,race and antiepileptic treatment process as well epileptic situation was record detailedly.GSTM1 and GSTT1 polymorphism were identified by using PCR amplification based on the presence and absence of a PCR amplification product.CAT polymorphism was identified using direct automated DNA sequencing.The correlation about antiepileptic drug-induced liver dysfunction and GST and CAT gene type was analyzed with statistical methods.Results:(1)Alanine transaminase(73.7%),aspartate aminotransferase(64.9%)andγ-glutamyltransferase(61.4%)were the main manifestations of liver dysfunction induced by antiepileptic drugs,among which ALT was the most abnormal.(2)Comparison of the experimental group and control group in the GST and CAT gene polymorphism:The P value of GSTM1,GSTT1 and CAT genotypes was analysied byc~2 test.There was no significant difference between the experimental group and the control group(P>0.05).(3)Comparison of abnormal liver function induced by each antiepileptic drug and control group in GST genotype:The results of GST genotype in experimental group and control group with each drug were compared.The P value of GSTM1 and GSTT1 genotypes was calculated byc~2 test.It was found that there was no significant correlation between GSTM1、GSTT1 genotype and abnormal liver function caused by VPA,OXC,LEV and CBZ individually(P>0.05).(4)Comparison of abnormal liver function induced by each antiepileptic drug and control group in CAT genotype:The results of CAT genotype in experimental group and control group with each drug were compared.The P value of CAT genotypes was calculated byc~2 test.It was found that there was no significant correlation between CAT genotype and abnormal liver function caused by VPA,OXC,LEV and CBZ individually(P>0.05).(5)To further test the correlation between GST and CAT genotypes and liver dysfunction:liver dysfunction is a dependent variable and GST and CAT genotypes are independent variables,and logistic regression analysis is carried out.Logistic regression showed that there was no significant correlation between GST and CAT genotypes and liver dysfunction induced by antiepileptic drugs(P>0.05).Conclusion:1.Alanine transaminase(73.7%),aspartate aminotransferase(64.9%)andγ-glutamyltransferase(61.4%)were the main manifestations of liver dysfunction induced by antiepileptic drugs,among which ALT was the most abnormal.2.Antiepileptic drugs-induced abnormal liver function is not significantly associated with GST and CAT gene polymorphism.
