Objectives:This study used high-throughput sequencing technology to analyze the differential expression and clinical significance of circular RNA in patients with acute cerebral infarction,in order to provide new ideas for the diagnosis and treatment of cerebral infarction.Methods:20 patients with acute cerebral infarction within 7 days of onset were collected as a case group?sex ratio 1:1?.The consultation time of these patients was from July 2017 to September 2017 and the location was at the Department of Neurology,China-Japan Union Hospital of Jilin University.The selected patients can be clearly diagnosed and meet the inclusion criteria.Another 10 males and 10 females with normal physical examination were selected as the normal control group,and the patient's plasma was collected.We used high-throughput sequencing to screen for differentially expressed circRNAs in the plasma of patients with acute cerebral infarction,volcanic maps and cluster maps show the difference in circRNAs,Target gene analysis software to predict target miRNAs that may interact with up-regulated and down-regulated circRNAs,functional analysis of differential genes was performed by genetic bioinformatics to explore the potential role of circRNAs in patients with acute cerebral infarction in order to find potential markers.We used real-time polymerase chain reaction?RT-qPCR?to validate the differentially expressed partial circRNAs.Results:1.In the plasma of patients with acute cerebral infarction,a total of 3764 circRNAs were detected in the specimens,volcanic maps and clustering results suggest that there are a large number of circRNAs in the experimental group compared with the control group.Among them,771 were up-regulated and 2993 were down-regulated.There were 18 circRNAs with significant differential expression,of which 3 were up-regulated and 15 were down-regulated.2.GO analysis showed that 130 items were obtained in patients with cerebral infarction?P<0.05?.Among them,the biological pathways?BP?of up-regulated the expression of circRNAs involving 24 items,and the down-regulated expression involved 40 items;the cellular components?CC?of up-regulated the expression of circRNAs involving 27 items,and the down-regulated expression involved 24 items;the molecular functions?MF?of up-regulated the expression of circRNAs involving 9 items,and the down-regulated expression involved 6 items.The results of KEGG pathway analysis showed that the up-regulated circRNAs had no enriched signaling pathway,while the down-regulated circRNAs had one signaling pathway for homologous recombination.In the annotation of circRNAs/miRNAs,the difference of 3764 circRNAs was predicted to bind to 5778 miRNAs,which listed significant differential expression of circRNAs predicted to bind to the top five.3.Six kinds of down-regulated circRNAs?hsacirc0001189,hsacirc0126525,hsacirc0001380,hsacirc0001558,hsacirc0001355,hsacirc0001445?were selected to design primers,which were verified by RT-qPCR and consistent with the sequencing results.Conclusion:1.The expression profile of circRNAs in patients with acute cerebral infarction has changed,and there are a large number of circRNAs that are up-regulated or down-regulated.2.GO analysis,KEGG pathway analysis,and miRNAs prediction of differentially expressed circRNAs generally suggest that circRNAs play an important role in patients with cerebral infarction.Circular RNA may be a new target for diagnosis and treatment of cerebral infarction.3.Six circRNAs were verified by RT-qPCR,which was consistent with the results of gene sequencing,providing clues for further study of cerebral infarction.
