Objective Family with sequence similarity 96,which conclude member A(FAM96A)and member B(FAM96B),are a kind of genes that were firstly found through comparing the genomes between humans and nematodes according to expressed sequence tag database of human.Recently researches have suggested that FAM96A and FAM96B were homologous proteins,both of them participate in tumorigenesis and progression as well as invasion and angiogenesis as potential cancer suppressor genes.FAM96A and FAM96B have a low expression in cancer tissue in human liver cancer and colon cancer,so they become a potential diagnosis mark and therapy target.Nevertheless,the role FAM96A and FAM96B played in gastric cancer haven't been reported yet.In this study,we investigate the expression of FAM96A and FAM96B in gastric cancer and explore the correlation between genetic expression and its clinical biological characteristics of gastric cancer.Furthermore,the proliferation as well as apoptosis effect of FAM96A and FAM96B overexpression on gastric cancer cell line SGC7901 are also evaluate by us.Methods Firstly,Immunohistochemistry was applied to examine the location of FAM96A and FAM96B protein in gastric cells.Western blot analysis was used to detect the FAM96A expression in 70 pairs of gastric cancer tissues and their adjacent normal tissues as well as FAM96B expression in 80 pairs of gastric cancer tissues and their adjacent normal tissues,all tissues mentioned above were surgically removed and pathologically confirmed in our hospital.SPSS was utilized to analyse the correlation between FAM96A and FAM96B's expression and their clinicopathological characteristics of gastric cancer.Furthermore,synthesized recombinant plasmid of pcDNA3.1-myc-FAM96A that containing the whole sequence of human FAM96A gene was transfected into SGC-7901 cells,the same method was used in the group of FAM96B.The proliferation of SGC-7901 cells was tested by MTT assay,Flow cytometry was employed to explore the apoptosis of cells.Results Immunohistochemistry uncovered that FAM96A protein was located in cytoplasma.Moreover,the positive expression rate of FAM96A in tumor adjacent tissues and gastric cancer tissues was 80%(56/70)and 39%(27/70)respectively(P<0.05).Western blot pointed that the relative expression levers of FAM96A protein in gastric cancer tissues was(0.45±0.15),conspicuously lower than(1.21±0.30)in adjacent tissues with the difference being statistically significant(t=11.994,P<0.01).There were significantly different between the amount of FAM96A protein expression and depth invasion,lymph node metastasis,distant metastasis,tumor node metastases(TNM)stage(P<0,01),but not in gender,age,tumor size(P>0.01).MTT assay indicated the cell proliferation's speed in FAM96A group was obviously lower than that in control group(P<0.05).Flow cytometry analysis showed the cell apoptotic rate in FAM96A group was higher than that in control group(P<0.05).Similarly,Immunohistochemistry indicated that FAM96B protein was situated in cytoplasma too.In addition,the positive expression rate of FAM96B in tumor adjacent tissues and gastric cancer tissues was 76%(62/80)and 45%(36/80)respectively(P<0.05)· The relative expression levers of FAM96B protein in gastric cancer showed in Western blot was(0.28±0.08),significantly lower than(1.71±0.29)in adjacent tissue with the difference being statistically significant(t=25.218,P<0.05).There were remarkably disparate between FAM96AB protein expressions and depth invasion,lymph node metastasis,distant metastasis,tumor node metastases(TNM)stage(P<0.05),but gender,age,tumor size were not involved(P>0.01).MTT assay indicated the cell proliferation's speed in FAM96B group was distinctly lower than that in control group(P<0.05).It has been proved that cell apoptotic rate in FAM96B group was higher than that in control group(P<0.05)according to Flow cytometry analysis.Conclusions The protein of FAM96A and FAM96B are both lower expressed in gastric cancer tissues significantly when compared with its adjacent tissues,the degree of which are closely correlated with the development,invasion and metastasis of gastric cancer.The overexpression of FAM96A and FAM96B could inhibit the SGC7901 cells' proliferation and induce its apoptosis likewise.So FAM96A and FAM96B may be an ideal biomarker that can apply on the diagnosis and prognosis prediction in patients with gastric cancer.
