Background and Object: micro RNA participates in a variety of pathological and physiological processes as an important component of noncoding RNA,including the start and development of tumors.After searching a series of articles,we come to a conclusion that Mi RNA-9-5p takes an important role in the regulation of human tumor process,especially to inhibit the malignant biological behavior of tumors.But the specific mechanism of how the tumors be regulated is far from fully elucidated.This is the reason why the article was written.We tried to explain clearly the mechanism how Mi RNA-9-5p affects the proliferation,metastasis and invasion of prostate cancer cells.Methods: The expression of mi R-9-5p in normal prostate epithelial cells R WPE-1,prostate cancer cells PC3 and DU145 was detected by using real-time PCR.And at the same time,the expression of mi R-9-5p and CXCR4 were detected by using real-time PCR.The Lenti-vrius carrying mi R-9-5p were used to infect PC3 cells to construct stable cell lines(PC3/mi R-9-5p),and the expression of mi R-9-5p in this cells were determined by using real-time PCR.Cell viability was assayed using a CCK8 kit according to the manufacturer's instruction at indicated time points.Migration and invasion were assayed using Transwell cell migration and invasion assay.Bioinformatics is used to predict the target genes of mi R-9-5p.The wild type(WT)and mutant type(Mut)luciferase reporter plasmids of target g ene 3'-UTR were constructed.The predicted results were verified by luciferase assay and real-time PCR.Results The expression of mi R-9-5p in normal prostate epithelial cells RWPE-1 was significantly higher than that in prostate cancer cells PC3 and DU145.And the expression of mi R-9-5p in normal tissues were also significantly higher than that inmiR-9-5 p was significantly inhibit cell migration and invasion of PC3 cells(P<0.01).CXCR4 could partially reverses the tumor suppressive effect of mir-9-5p.Bioinformatics prediction indicates that mi R-9-5p can target the 3'-UTR of CXCR4.Luciferase assay and real-time PCR show that mi R-9-5p can target the 3'-UTR of CXCR4 and then inhibit the expression of CXCR4 m RNA.tumor tissues.Then the level of mi R-9-5p in PC3/mi R-9-5p cells was significantly higher than that in PC3 cells(P<0.01)and control cells(P C3/mi R-NC)(P<0.01).Cell viability assay showed that compared with PC3 group(P<0.01),the cell viability of PC3/mi R-9-5p group decreased significantly at 72 h and 96 h.Overexpression ofConclusion: We have identified that mi R-9-5p inhibits cell proliferation,migration and invasion in prostate cancer cells,and the mechanism may be achieved by targeting the 3'-UTR of CXCR4.
