OBJECTIVE:To investigate the expression of RNA and protein in gastric cancer tissues and adjacent tissues of m6A methyltransferase KIAA1429,and to influence the proliferation,migration and invasion of gastric cancer cells.Methods:The mRNA and protein expressions of KIAA1429 gene in gastric cancer tissues and adjacent tissues were detected by Quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR)and Western blotting.Human gastric cancer cell lines SGC7901 and MGC803 were cultured in vitro,and lentiviral transfection technology was used to construct KIAA1429-interfering gastric cancer stably transfected cell lines by shKIAA1429-1 and shKIAA1429-2,and CCK8 experiments and plate clone formation experiments and scratches were performed.Healing experiments and Transwell migration and invasion experiments were carried out to investigate the effects of KIAA1429 on the proliferation,clonality,migration and invasion ability of human gastric cancer cell lines SGC7901 and MGC803.Finally,subcutaneous tumor formation experiments in nude mice were further confirmed in vivo.Results:Compared with normal tissues adjacent to gastric cancer,the mRNA and protein expression levels of KIAA1429 in 20 cases of gastric cancer were higher(P<0.05).In gastric cancer cell lines SGC7901 and MGC803,the proliferation rate of cells after interference with KIAA1429 was significantly lower than that of the control group,and the monoclonal formation rate of cells after interference with KIAA1429 was significantly lower than that of the control group.In the Traswell migration and invasion experiments,compared with the control group,the interference with KIAA1429 expression significantly reduced the migration and invasion of gastric cancer cells,and the migration rate decreased(P<0.05).In the subcutaneous tumor formation experiment in nude mice,the subcutaneous tumor growth rate of nude mice was slower than that of the control group,and the volume and quality of the final tumor were smaller than the control group(P<0.05).Conclusions:KIAA1429 gene is highly expressed in gastric cancer and could promote proliferation,migration and invasion of gastric cancer cells.The results of this study can provide clues for further research on the in-depth mechanism and clinical application of KIAA1429 in the development of gastric cancer.
