Background and objective Primary dysmenorrhea is one of the common diseases in gynecology.Severe dysmenorrhea will bring about a lot of convenience,resulting in absence of classes or work.The therapeutic effect of traditional Chinese medicine for primary dysmenorrhea has been recognized clinically.However,Chinese medicine was featured by multi-molecule and multi-target,and its action mechanism still needs to be further studied.Take Tongjingtang as an example,the objective of study dissects the action mechanism of Tongjingtang through systems pharmacology method.Method The potential active compounds and targets of Tongjingtang were excavated by the Traditional Chinese Medicine Systems Pharmacology analysis platform(TCMSP).Then acquired targets were screen out by Uniprot and DAVID database for further analyze diseases and pathways related to primary dysmenorrhea by CTD and KEGG database.The networks and integrated pathways were established by Cytoscape and Visio software for analyzing the action mechanism of treating primary dysmenorrhea with Tongjingtang.Result The 93 active compounds and relative 133 target proteins of Tongjingtang were screened out for building compound-target and compound-target-disease networks.The network analysis shows the active material basis of the compound and the links between active molecules,targets and diseases from a holistic and systematic point of view.The target protein-related pathways mainly refer to regulate the uterine blood vessels,smooth muscle contraction and relaxation,uterine blood perfusion,inflammatory response and sex hormone levels of patients with primary dysmenorrhea.Conclusion Through the systems pharmacology method,the study reveal the molecular mechanism of Tongjingtang by molecular interaction between multiple targets and more and multiple pathways regulation for the treatment for primary dysmenorrhea,which provide reference for the modernization of traditional Chinese medicine research.
