Objective:To explore the Dihuangyinzi intervention in Alzheimer's disease?AD?and Parkinson disease?PD?and the effect mechanism of kidney deficiency rat model,reveals Dihuangyinzi intervention AD and kidney deficiency of PD same syndrome in different diseases of biological mechanism.Methods:?1?Through reviewing the theory of traditional medicine and modern medicine in the literature of AD,PD,pathogenesis and pathogenesis,combing the relationship among the factors of disease,syndrome,syndrome,and clarifying the reason and basis for the treatment of AD and PD with kidney deficiency syndrome.?2?Replicating disease syndrome combined with animal model:the animal model of kidney deficiency syndrome in TCM was simulated with hydrocortisone solution,then the animal model of AD disease was copied with A?25-355-35 peptide fragment from the CA1 region of the hippocampus,and rotenone was injected subcutaneously on the back of the neck to replicate the animal model of PD disease,and AD was prepared to be AD respectively.The model of PD disease combined with syndrome was established.After the end of modeling,the intervention model of combination of AD and PD was established by Dihuang Decoction.?3?The behavioral changes,morphological changes,HPA axis function changes,the changes in the HPA axis function,the changes in the antioxidant water level of the brain tissue,the changes of the UPS function and the intervention of the Radix Rehmanniae Decoction were observed by the behavioral test,ELISA,HE staining,Western blot and RT-PCR transcription polymerase chain reaction.Results:1.theoretical study:?1?modern doctors believe that both AD and PD belong to the aging disease.The disease is in the brain and the deficiency of the kidney is the common basis of the disease."Kidney deficiency"is the common syndrome factor,which belongs to the category of the same syndrome in traditional Chinese medicine.The kidney nourishing recipe,Rehmannia glutinosa,has the function of filling the marrow,nourishing kidney yin,tonifying kidney yang,reducing phlegm and inducing resuscitation,and can be used for treating AD and PD deficiency of kidney.?2?Modern medicine thinks that the UPS function decline caused by aging is one of the common pathogenesis of AD and PD,and the modern Chinese and Western Medicine on renal parenchyma has been defined as the physiological kidney deficiency.Therefore,the physiological kidney deficiency is closely related to the decline of the function of the ubiquitin proteasome system.2.experimental study:?1?the behavioral test results:AD model rats with memory space exploration ability of serious damage,Dihuangyinzi can improve the AD of the intervention group rats learning ability;reduce PD rat limbs muscle strength and balance ability,Dihuangyinzi can improve the PD intervention group rats limb muscle strength and balance ability.?2?HPA axis function test results:AD and PD rats serum CTH,ACTH,CORT concentration increased,showed that the HPA axis function deficiency hyperthyroidism,kidney deficiency syndrome model was constructed successfully,with Dihuangyinzi intervention,AD,PD,ACTH,CTH concentration of intervention group rats serum CORT decreased the table Ming Dihuangyinzi can regulate HPA axis function deficiency hyperthyroidism,has preventive effects on AD and PD of kidney deficiency syndrome model.?3?the results of Lipo,AD,MAO:PD model group rat brain homogenates of MAO,Lipo concentration increased significantly,showed that brain tissue decreased antioxidant levels,that kidney aging model induced by the successful Dihuangyinzi intervention AD,PD intervention MAO and Lipo in brain tissue of rats decreased,suggesting that Dihuangyinzi the intervention group can improve rat brain antioxidant levels,inhibit rat brain aging,has certain therapeutic effect on AD and PD model combined with kidney deficiency syndrome.?4?the results of HE staining:hippocampal cell layers in AD model group rats decreased,cell vacuolar degeneration significantly,Dihuangyinzi can increase the intervention of AD number of nerve cells in the hippocampus of rats,reduce cell vacuolar degeneration;PD rat mesencephalic substantia nigra DA neurons reduced the number of glial cells increased.The PD intervention group,Dihuangyinzi rat mesencephalic substantia nigra DA neurons with the increase in the number of glial cells decreased in number.?5?Western-blot in the detection of UPS expression in brain tissue of rats in the model group AD,PD Ub,E1,UCH-L1 and 20S proteasome protein decreased,showed UPS dysfunction,with Dihuangyinzi intervention,AD and PD on the expression of brain tissue in rats of Ub,E1,UCH-L1 and 20S proteasome egg white increased,suggesting that dhyz can restore the degradation function model of AD/PD groups.?6?RT-PCR in the detection of UPS expression in brain tissue of rats in the model group AD,PD Ub,E1,UCH-L1 and 20S proteasome mRNA decreased,indicating that UPS function decreased by Dihuangyinzi intervention,AD and PD treatment group rat brain tissue in Ub,E1,UCH-L1 and 20S proteasome mRNA the table was increased,indicated that the degradation function Dihuangyinzi can restore AD/PD group model.Conclusion:1.the combination model of AD syndrome with AD syndrome could be successfully replicated by intragastric intragastric injection of hydrocortisone solution in Wistar rats and A beta 25-35 peptide fragment from the hippocampus of the hippocampus.2.the combination model of PD kidney deficiency syndrome can be
