Objective:Eukaryotic translation elongation factor 1A1(eEF1A1)is an evolutionarily conserved elongation factor protein,which is involved in the proliferation of tumor cells and highly expressed in a variety of tumor cells.In addition,it has been confirmed that the cell cycle is related to the proliferation of tumor cells,and the Cyclin D1 has close connections with the G1 phase of the cell cycle.This dissertation mainly discusses the relationship between eEF1A1 and cyclin D1.Methods:1.Western blot and rt-pcr were used to detect the protein and mRNA expression of eEF1A1 in HepG2,Huh7,HCCLM3,MHCC97 H,Hep3B and hl-7702 in immortalized hepatocytes.2.With the construction of stable low expression eEF1A1 hepatocellular carcinoma cell lines by lentivirus,the proliferation and cycle of tumor cells in stable low expression eEF1A1 hepatocellular carcinoma cell lines were observed by EDU assay and flow cytometry;3.After changing the expression level of eEF1A1 in hepatocellular carcinoma cells,the changes of protein and mRNA of STAT1 and cyclin D1 were observed;4.STAT1 and cyclin D1 were over-expressed in stable and low-expression eEF1A1 hepatocellular carcinoma cell lines respectivelyto observe the proliferation of hepatocellular carcinoma cells.Results:1.Western blot and real-time fluorescence quantitative analysis(RT-PCR)showed that the expression of eEF1A1 in hepatocellular carcinoma cell lines is higher than that in normal hepatocytes..2.In HCCLM3 hepatocellular carcinoma cell line with stable and low expression of eEF1A1,cell cycle arrest is in G1 phase,and the proliferation ability of hepatocellular carcinoma cells decreases.3.By down-regulating the expression of eEF1A1 in HCCLM3 hepatocellular carcinoma cell lines,the protein and mRNA levels of STAT1 and cyclin D1 also descend.4.EEF1A1 regulates the cell cycle of hepatocellular carcinoma by controlling the expression of cyclin D1 through STAT1,thus affecting cell proliferation..Conclusion:In hepatocellular carcinoma cells,when down-regulating the expression of eEF1A1,the expression of STAT1 and cyclin D1 decreased significantly,then cell cycle is blocked in G1 phase,and the proliferation ability of hepatocellular carcinoma cells decreases.However,down-regulation of STAT1 does not affect the change of eEF1A1,but only decreases the expression of cyclin D1.These results suggest that eEF1A1 can control the proliferation of tumor cells through STAT1-cyclin D1 axis.
