Objective:The objective of this study is to investigate whether there were differences in the plasma levels of miR-202 in CHD and healthy people,and to analyze the relationship between mir-202 and the risk of coronary heart disease。Methods:This study included 81 patients(case group)with coronary heart disease who visited the department of cardiovascular medicine of the Second Affiliated Hospital of Nanchang University from November 2016 to August 2017,and 81 patients(control group)who underwent routine physical examination in the Second Affiliated Hospital of Nanchang University during the same period 。 The two groups were matched by gender(males accounted for 79% in the control group and 74.1% in the case group,P=0.458)and age(control group 63.81±9.17,case group 66.12±8.90,P=0.106)composition。Procedure:5ml venous blood of the fasting elbow of the subjects was collected to obtain peripheral blood samples for RNA extraction 。Real-time fluorescent quantitative PCR(qRT-PCR)is used to obtain the expression of miR-202 levels,and made by U6,through calculating-Δ Ct relative expression of miR-202。General information of all subjects(including age,sex,height,weight,resting blood pressure,history of smoking,history of drinking,history of hypertension,history of diabetes,family history of coronary heart disease,etc)and Clinical test results(including routine hematological examination,liver and kidney function test,blood lipid and fasting blood-glucose,etc)。Results:(1)The plasma level of miR-202 in the CHD group was significantly higher than the control group(-1.42±2.04 vs-2.67±1.95),and the difference was statistically significant(P < 0.001)。(2)Adjusted for age and sex,history of smoking,history of alcohol consumption,history of hypertension,history of diabetes,history of triglycerides,and found that elevated miR-202 level was associated with increased risk of coronaryheart disease(Adjust OR 1.37,95%CI 1.11-1.70,P = 0.004)。(3)MiR-202 level was negatively correlated with platelet width(r=-0.348,P=0.002)and mean platelet volume(r=-0.29,P=0.01)in patients with coron-ary heart disease.(4)The area under the ROC curve of the baseline model was 0.84(95% Cl0.78-0.90,P < 0.001)similar to that of the miR-202 combined prediction model 0.88(95% CI 0.82-0.93,P < 0.001)(P=0.147)。Mir-202 was able to reclassify patients with NRI of 50.5%(95%CI 20.3-80.7,P=0.0015)and IDI of 0.04(95% CI 0.01-0.07,P=0.0096).Conclusion:(1)Elevated plasma miR-202 expression was associated with an increased risk of coronary heart disease。(2)Our resecach suggest that plasma miR-202 levels may be a potential noninvasive biomarker for the diagnosis of coronary heart disease.
