Artichoke(Cynara scolumus L.)is a genus of Cynara L.in the Comopsite family,in the folk,its extract,as a medicinal,has the functions of promoting digestion,liver protection,anti-cancer,anti-HIV,cholagogic,anti-inflammatory and antibacterial function,and inhibition of cholesterol biosynthesis.Studies have shown that its functional components are flavonoids,polyphenols,terpenoids and their sesquiterpenoids(especially guaiacols sesquiterpenes and their lactones).The functional components of artichokes vary with different species.Therefore,this study used artichoke"Lorca"as raw material to study the separation and purification of its functional components and the structural identification of the pured peaks obtained.The specific methods and conclusions are as follows:(1)METHOD The artichoke head powder was extracted with 70%ethanol for 72 h,and the ratio of material to liquid was 1:10(m/v).The crude extract was separated with a macroporous resin AB-8 and eluted with different concentrations of ethanol as eluents.Fraction A27-29 was purified by RP-C18 column chromatography,and the purified parts were subjected to high performance liquid chromatography detection and preparation.RESULT A total of 12 pured peaks were obtained by purification and preparation.They are:H-01 and H-02 from part C16-19;H-03,H-04 and H-05 from part C11-15;H-06,H-07,H-08,H-09 and H-11 from part C5-10;and H-10 and H-12 from the Fr.2(C25-29 combined with A35-36).Among them,H-01,H-05,and H-09 were the same compounds by HPLC retention time,UV and LC-MS.(2)METHOD The 8 pure partions were analyzed by LC-QTOF-MS and NMR(1H-NMR?13C-NMR?H-H COSY?HMBC?HSQC).RESULT The structures of 8 compounds were determined by data of analysis and literature.They are luteolin 7-O-?-D-glucoside(compound 2),cynaratriol(compound 3),grosheimol(compound 4 and 8),luteolin 7-O-?-L-rhamnosyl(1-6)-?-D-glucoside(compound 5),cynarinin B(compound 6),azuleno[4,5-b]furan-2(3H)-one,3-(chloromethyl)decahydro-3,8-dihydroxy-9-methyl-6-methylene-,(3S,3aR,6aR,8S,9S,9aR,9bR)(compound 7a),isolipidiol(compound 7b),and cynaropicrin(compound 10).Among them,2 are flavonoids and 6 are guaiane-type sesquiterpene lactones(H-07 is a mixture of H-07a and H-07b),and H-07a is a new compound.
