Objective:To observe the efficacy of pulsed electrical stimulation combined with curcumin in the treatment of melanoma in the dorsal skin of mice.Methods:B16-F10 mouse melanoma cells were injected into the dorsal skin of C57BL/6 mice to make a dorsal melanoma model.The transplanted mice modeled at the same time were randomly divided into 4 groups: control group and electrical stimulation group.,curcumin group,joint group.The electrical pulse pulse width was set to 5 ms,the frequency was 20 Hz,and the stimulation voltage was 20 V/cm.The mice in the electrical stimulation group received electrical stimulation for 7 consecutive days(30min/d).The control mice were treated in the same manner as before,and the stimulation frequency was set to 0 Hz.The pulsed electric field was used to treat melanoma independently or in combination with curcumin.The mouse foot was placed in a foot volume measuring device and observed daily to record the subcutaneous tumor growth of the mouse.If a blue-black nodule with a shortest diameter of 0.5-0.8 cm was observed under the skin of the mouse,the mouse xenograft model was successfully established.Hematoxylin-eosin(H-E)staining was used to observe the growth of tumor cells in the skin.Apoptosis assay was used to observe the effect of pulsed electrical stimulation alone or in combination with curcumin on tumor apoptosis.Results:The skin melanoma model was successfully produced by injection of B16-F10 melanoma cells into the back of the foot;pulsed electrical stimulation alone or in combination with curcumin has the effect of suppressing the enlargement of melanoma,and the combination therapy is more effective,and the combined treatment group tumor cells The number of apoptosis is significantly increased.The tumor volume inhibition rate was statistically significant(P<0.05),and the combined tumor volume inhibition rate was the highest,followedby the second electrical stimulation group;the tumor volume growth rate was not statistically significant except for the control group and the curcumin group(P>0.05),the differences between the other groups were statistically significant(P<0.05),and the combined tumor volume growth rate was the lowest,followed by the electrical stimulation group,the control group,the curcumin tumor volume was equivalent,and the groups were black.Under the light microscope of TUNEL staining,the apoptotic cells of the melanoma cells in the control group were less and scattered.The apoptotic cells in the pulsed electrical stimulation group increased compared with the control group,which was statistically significant.Pulse electrical stimulation combined with turmeric In the treatment group,the tumor parenchyma density was decreased,and some morphological changes of nuclear pyknosis and fragmentation were observed,and a large number of apoptotic cells were observed.Conclusion:The dorsal melanoma model is easy to observe the morphology and volume of the tumor.It has certain advantages.Pulse electrical stimulation combined with curcumin can inhibit the growth of melanoma by causing apoptosis,which is a more effective treatment for melanoma.
