| Objective: This study aimed to explore the effect of oats and tea polyphenols on metabolic syndrome(MS)and mild persistent inflammatory state of MS,as well as exploring the relationship between peroxisome proliferator-activated receptor-γ and MS components.Methods: Eight-week-old male SD rats(48)were randomly divided into two groups,the normal group(Control group,basal diet)and the model group(High-fat diet and 15% fructose water),and both of groups were fed for 10 weeks.All of experimental rats were weighed weekly,and the relevant indicators(obesity index,insulin resistance-related index,blood lipids and blood pressure)were detected every 4 weeks,and the inflammation-related indicators were measured at the 10 th week.Meanwhile,whether the model of MS rats was made successfully was judged by the diagnostic criteria.According to the principle of factorial design,MS rats were randomly divided into 5 groups: model control group(MS control group),oat intervention group(Oat group),tea polyphenol intervention group(TP group),oatmeal + tea polyphenols,Group(OT group)and oatmeal + tea polyphenol + PPAR-γ inhibitor group(OTP group).Oatmeal feed is a kind of special high-fat diet whose some ingredients,wheat and corn,are all instead of oats.Tea polyphenols were added to rat drinking water at 400mg/kg.The PPAR-γ inhibitor T0070907 was given rats once every other day at a dose of 1 mg/kg body weight by intraperitoneally injected.Six groups of rats were fed for 12 weeks.The general condition of the rats was observed during the feeding period,and the body weight of the rats was weighed every week,and the relevant indexes were detected every 4 weeks.After 12 weeks of intervention,the rats in each group were killed immediately after anaesthesia,and then they were uniformly took out varioustissues required for this study to be conducted some related tests.Results: 1.The rate of rats becoming MS was 87.5%.2.Effect of oats and tea polyphenols on obesity index of MS rats: 1)Body weight: During the intervention period,Oat group,OT group and OTP group decreased,and the other groups showed an increasing trend;2)Waist circumference: Oat group,OT group and OTP group stopped growing,and there was a slow decrease trend,while other groups showed a growth trend;3)Lee’s index: OT group decreased,MS Control group increased,and the trend of other groups before and after intervention was not significant;4)Visceral fat: The visceral fat,liver weight and visceral fat to body weight ratio,Oat group,TP group and OT group are lower than MS Control group and OTP group.Oats and tea polyphenols have an interaction in reducing visceral fat,heart,liver weight,visceral fat to body weight ratio and liver to body weight ratio in rats.3.Effects of oats and polyphenols on IR related indexes in MS rats: 1)Fasting blood sugar: The reduction trend of Oat group,TP group,OT group and OTP group was significantly higher than that of MS Control group;2)Fasting insulin: Oat group,TP group,OT group and OTP group all showed a decreasing trend,while other groups showed an increasing trend;3)Homeostatic model of insulin resistance index: Oat group,TP group,OT group and OTP group all showed a decreasing trend,while the other groups remained relatively stable.Oats and tea polyphenols have interactions in reducing Fasting insulin and HOMA-IR.4.Effects of oats and tea polyphenols on blood lipids in MS rats: 1)High density lipoprotein cholesterol(HDL-C): Oat group,TP group and OT group increased,while the other groups remained relatively stable;2)Triglyceride(TG): Oat group,TP group and OT group were reduced,and the other groups remained relatively stable.Oats and tea polyphenols reduced TG interaction,while elevated HDL-C had no interaction.5.Effects of oats and tea polyphenols on blood pressure of MS rats: 1)Systolic blood pressure: Oat group,TP group,OT group and OTP group all showed a decreasing trend,while other groups showed an upward trend;2)Diastolic blood pressure: TP group andthe OT group decreased,and the other groups increased.Oats have no interaction with tea polyphenols.6.Effects of oats and tea polyphenols on inflammatory status in MS rats: 1)IL-6,TNF-α,CRP and MCP-1: Oat group,TP group and OT group were reduced,and the other groups remained stable;2)ADPN: Oat group,TP group and OT group were elevated,and the other groups remained stable.Oats and tea polyphenols have an interaction in improving the levels of inflammatory factors.7.The effect of oats and tea polyphenols on the expression of PPAR-γ mRNA in adipose tissue of MS rats: Oat group and OT group are higher than OTP group and MS Control group.Oats have no interaction with tea polyphenols.8.Effects of oats and tea polyphenols on the tissue structure of MS rats: There were no abnormalities in the heart,kidney and pancreas of each group;the liver showed different severity of steatosis.Conclusions: Increasing the intake of refined sugars such as fructose can promote the occurrence and development of MS in rats.Adding oats and tea polyphenols together in the feed has better effect on controlling blood sugar and lose weight in rats.Oats can improve MS obesity and lipid metabolism disorder by up-regulating PPAR-γ expression,and this effect is superior to tea polyphenols. |
