Morindae Officinalis Radix(MO-Radix)is the dry root of Morinda officinalis How.,one of the best-known herbs in southern China.It has the function of treating impotence and female infertility,menstrual disorders,cold and pain in the lower abdomen,rheumatoid arthritis and osteoporosis[1].In recent years,with the improvement of living standards,more and more people are paying attention to diet supplements.MO-Radix is widely used as a nutrient supplement[2].MO-Radix are mainly distributed in four provinces of Guangdong,Guangxi,Fujian and Hainan.The genuine producing area is the Deqing and Gaoyao areas in Zhaoqing,Guangdong.But so far,there is no effective way to distinguish the MO-Radix from different geographical origins.Therefore,to process a reliable method to determinate the Geographical origins area of MO-Radix is important to guarantee the efficacy of clinical application,protect the brand characteristics and promote the development of the genuine area.Objective:This paper systematically studied the origin determination of MO-Radix,and discussed the discrepancy of MO-Radix from various geographical origins and the contribution of different indicators to the discrepancy.Established a discriminant model which can quickly and effectively distinguish the Geographical origin of MO-Radix.Method:1.Stable isotope measurement:Ultrafine powder was obtained through 200 mesh sieves.After burning by EA,the sample was converted into gas to be measured.Then it was detected by IR-MS.The results were corrected by two-point correction method.2.Mineral elements analyzed:The samples were digested by microwave digestion apparatus.And screened high response value,low consumption of reagents and short time were by orthogonal test,and ICP-MS was used to measure the repeatability and·0-=;'/precision of the method.3.Determination of oligosaccharide:Samples were extracted in boiling water with mobile phase for 30 minutes and filtrated.Analyses were performed using a C18-PFP reverse chromatography column and the temperature was maintained at 30 ?.The mobile phase consisted of methanol(A)and water(B)(3:97)and the flow rate is 0.8 ml/min.The spectra were recorded by ELSD and drift tube temperature is 110 ? nitrogen flow rate is 2 L/min,injection volume is 10?l;collection time is 12 min.4.Using LDA method to screen factors and establish a linear discriminant model with fewer indices and higher discriminant accuracy.Result:1.It is inaccurate to distinguish eight areas of MO-Radix by stable isotopes.The rate of divided the eight sites into three areas is 86.9%.It shows that MO-Radix in different provinces can be distinguished by stable isotopes.2.Orthogonal design was used to optimize the microwave digestion method of MO-Radix.The optimal solution was 5 ml nitric acid,150 C holding tine 10 min in the first step,180? holding tine and 20 min in the second step.22 mineral elements in MO-Radix were detected by ICP-MS.An effective discriminant model of MO-Radix could be established by using these mineral elements,and the accuracy classification rate was 98.4%.3.The identification accuracy to distinguish areas of MO-Radix by peak area of oligosaccharide is only 75.4%.The oligosaccharide of MO-Radix from different producing areas are similar,but the content is different.Among these areas,the content of oligosaccharides in MO-Radix from Deqing is the highest.4.Combined with stable isotopes,mineral elements and oligosaccharide,12 specific indicators were screened by LDA,including 1 stable isotope,10 mineral elements and 1 oligosaccharide.The accuracy rate of the model is 98.4%,which has practical application value.Gonclusion:1.The stable isotopes,mineral elements and oligosaccharide of MO-Radix have significant difference among various geographical origins.It is feasible to trace the geographical origin of MO-Radix.2.Established a discriminant model of MO-Radix origin based on the combination of stable isotopes,mineral elements and oligosaccharide.There are 12 indexes needed,and the classification rate is 98.4%.3.Stable isotopes and oligosaccharide lack of accuracy in discriminate the geographical origins of MO-Radix,and mineral elements is better than the two indicators.4.The content of oligosaccharide in MO-Radix from different geographical origins was different.The quality of MO-Radix in Deqing area is the best if oligosaccharides are used as the criterion.
