| Objective: The expression of MNK2(MAPK signal-integrating kinases-2)was investigated with significance and clinical in gastric cancer.Methods: Fresh tumor tissue and corresponding paracancerous tissue specimens from 16 patients with gastric cancer who underwent surgical treatment in department of gastrointestinal surgery of The First Affiliated Hospital of University of South China in 2017 were collected;Paraffin specimens from 120 patients who underwent radical gastrectomy at The First Fffiliated Hospital of University of Ssouth China from December2010 to December 2013 were collected and included in this study group.Using the RT-PCR detection of 16 cases of gastric cancer tissue and suitable for fresh tissue adjacent to carcinoma MNK2 m RNA expression;Using Western Blot detection of 16 cases of gastric cancer tissue and suitable for fresh tissue adjacent to carcinoma MNK2 protein expression;IHC were used to detect 120 cases of paraffin tissue specimens MNK2 protein expression in patients with gastric cancer,and to analyze its expression and clinical pathological features of correlation(age,gender,whether the leaching of size,TNM stages,differentiation degree,infiltration depth,lymph node positive and tissue types,etc.)and its relationship with clinical prognosis.Results: RT-PCR test results show that fresh gastric carcinoma tissue MNK2 m RNA expression is significantly higher than the corresponding tissue adjacent to carcinoma(P < 0.05);Western Blot results showed that MNK2 protein was expressed in both fresh gastric cancer tissues and the corresponding paracancer tissues,and its expression in the cancer tissues was significantly higher than that in the corresponding paracancer tissues(P < 0.05).IHC test results showed that high MNK2 protein in 120 cases of gastric cancer paraffin tissue expression in 67 cases(55.8%),lower expression of 53 cases(44.2%),further analysis showed that MNK2 protein expression level is closely related to the tumor infiltration depth(P < 0.05),but with the patient age,tumor size,type of tissue differentiation,lymph node metastasis,distant metastasis,TNM staging no significant correlation(P > 0.05);Further follow-up showed that the survival rate of the low expression group of MNK2 protein was significantly higher than that of the high expression group(P=0.0102).Conculusion:1.MNK2 m RNA is significantly higher in gastric cancer tissues than in adjacent cancer tissues.2.The expression level of MNK2 protein was significantly higher in fresh gastric cancer tissues than in adjacent tissues.3.MNK2 is highly expressed in paraffin tissues of gastric cancer and is related to the depth of tumor invasion;4.High expression of MNK2 is associated with poor prognosis of patients and can be used as a potential therapeutic target for gastric cancer. |
