Radix puerariae is the dried root of the legume plant kudzu which can be used as both chinese medicine and food.Pueraria is rich in flavonoids,triterpenoid saponins,daidzein and other medicinal ingredients,as well as protein,dietary fiber and minerals?Fe,Zn,Ca,P,K,etc.?and other nutrients.Pueraria has the effects of relieving muscle fever,producing saliva and quenching thirst.It also has special effects on hypertension,coronary heart disease,angina pectoris,diabetes,and tumor.In this experiment,Pueraria lobata was used as raw material,and the main components?puerarin,starch,protein,dietary fiber,trace elements,etc.?of Pueraria lobata were tested by corresponding methods.The results of ethanol reflux extraction and ultrasound-assisted extraction were compared for selecting the best extraction process.The extraction was subjected to acid hydrolysis to increase the extraction ratio of puerarin.The crude extraction of puerarin was separated and purified by anion exchange resin.The experimental results are as follows:?1?Determination of Puerarin,Starch,Dietary Fiber,Protein and Mineral Elements in Radix Puerariae from different areas of Jiangxi Province by High Performance Liquid Chromatography,Enzymatic Hydrolysis,Kjeldahl Analyzer,Enzyme Gravimetry,Atomic Fluorescence Spectrometry and Atomic Absorption Spectrometry content.The results show that the highest content of puerarin is Pueraria lobata from Shicheng county.The highest protein content is Pueraria lobata from Yongxin county.The highest starch content is Radix Puerariae from lengmeng Town of Hengfeng couty.The content of dietary fiber in Pueraria lobata from different areas has no significant difference,and its heavy metal elements content is in line with the relevant standards in the relevant national regulations.?2?Ethanol reflux method and ultrasound-assisted extraction method were respectively used to extract puerarin from pueraria,and the content of puerarin was detected by high performance liquid chromatography.The effects of ethanol concentration,reflux time,reflux temperature,ultrasonic time,ultrasonic temperature and solid-liquid ratio on the yield of puerarin were investigated.The ethanol refluxing process was as follows:ethanol concentration was 60%,reflux time was 2 h,reflux temperature was70?,and the ratio of material to liquid was 1:7 g/mL.At this time,the yield of puerarin was the highest,being 1.19%.The ultrasonic assisted extraction method was as follows:ethanol concentration was 70%,ultrasonic time was 20 min,ultrasonic temperature was50?,and the ratio of material to liquid was 1:10 g/mL.At this time,puerarin had the highest yield of 1.23%.Comparing the two methods,it was found that the ultrasonic assisted extraction took less time,costed less,and the yield was higher than the ethanol reflux method.Therefore,the ultrasound-assisted extraction method has a better extraction effect.?3?The effects of hydrochloric acid concentration,hydrolysis time and hydrolysis temperature on the acid hydrolysis of crude extract of puerarin were investigated by single factor experiment.Then the optimum conditions of hydrolysis were determined by L9?33?orthogonal test.The results showed that the best condition of puerarin acid hydrolysis is as follows:the concentration of hydrochloric acid for 3 mol/L,hydrolysis time of 5 h,hydrolysis temperature is 70?,at this point,the puerarin extraction yield increased by16.46%.?4?The effects of anion exchange resin mass,adsorption time,adsorption temperature and pH on the adsorption of puerarin were analyzed by single factor analysis.At the same time,the desorption experiment was carried out to analyze the effects of desorption solution concentration,desorption time,desorption temperature and pH value of desorption solution on the desorption of puerarin.The effect of the adsorption process and the desorption process was optimized by orthogonal test.The results showed that the adsorption ratio of puerarin was the best when the puerarin was adsorbed by the 4g resin which was 40?and pH 6 for 12 h.The adsorption ratio was 75.62%;Puerarin was eluted in a 70%eluate with a pH of 7 and a temperature of 30?for 7 h.The desorption ratio was the highest at 80.54%.
