| Objective:Modern physicians often recognize depression from “depression syndrome”,emphasizing“Stagnation of liver Qi” as its basic pathogenesis.However,our previous studies have shown that many typical clinical manifestations of depression patients,such as depression,laziness,fatigue,thinking,insomnia,despair,and even the occurrence of pain,suicide,are difficult to fully explain the pathogenesis of “Stagnation of liver Qi”,and the treatment of depression based on syndrome differentiation of “Stagnation of liver Qi” has not achieved the desired effect.Professor Ding Yuanqing has done a lot of literature research and clinical research.Professor Ding Yuanqing,on the basis of a large number of literature research and clinical research,combines the relevant knowledge of doctors of past dynasties,according to The Theory of “Yang-Qi nourishes spirit and tendons in essence and tenderness”,Put forward “Yang yu shen tui” as the Innovative Pathogenesis of Depression.Taking “Rouse Yang-Qi”as the basic treatment method,Cooperate with the method of nourishing Ying-Qi and Shen,and creating Clinical Experience Prescription “Qiang Zhi Fang”.This paper intends to demonstrate the reliability and rationality of the pathogenesis theory of “Yang yu shen tui” by means of animal experiments.Methods: Sixty healthy male Wistar rats of SPF grade were randomly divided into six groups according to open-field and sugar-water data after one week of adaptive feeding: normal group,model group,Chinese medicine treatment group(high-dose Qiang Zhi Fang group,low-dose Qiang Zhi Fang group),Chinese medicine control group(Chaihu Shugan San group)and Western medicine control group(fluoxetine group).The route of administration was intragastric administration once a day.Chronic mild stress(CMS)was used to replicate the depression rat model.The physiological metabolism,loss of interest,exercise and exploratory behavior of rats in each group were measured by body mass,sugar preference coefficient and open field experiment.The content of c AMP was detected by ELISA.The expression of PKA,CREB and BDNF was detected by RT-PCR and Western-Blot.All the data were analyzed by Graph Pad Prism 5 statistical software and LSD method.The results were expressed as (?)±sd,P<0.05.Results: Behavioral test results: Compared with the normal group,the body weight,sugar intake and total distance in open field of model group rats were significantly decreased(P<0.001);compared with model group rats,the body weight,sugar intake and total distance in open field of model group rats were significantly increased(P<0.01)after the intervention of Qiang Zhi Fang high dose,Qiang Zhi Fang low dose,Chaihu Shugan San and fluoxetine.However,there was no significant difference among the treatment groups(P>0.05).2.ELISA method was used to detect c AMP in hippocampus of rats.The content of cAMP in model group was significantly lower than that in normal group(P<0.01);Compared with model group,the content of c AMP in Qiang Zhi Fang low dose group,Chaihu Shugan San group and fluoxetine group was significantly higher(P<0.01),and that in Qiang Zhi Fang high dose group was significantly higher(P<0.05).There was no significant difference among the treatment groups(P>0.05).3.Western-blot method was used to detect the expression of PKA,P-CREB and BDNF in hippocampus of rats.Compared with normal rats,the expression of PKA,P-CREB and BDNF in hippocampus of rats in model group decreased significantly(P<0.001);Compared with model group,the expression of PKA,P-CREB and BDNF in Qiang Zhi Fang low dose group,Chaihu Shugan San group and fluoxetine group increased significantly(P<0.01).Qiang Zhi Fang high dose group also showed an increasing trend(P<0.05),and there was no significant difference among the treatment groups(P>0.05).4.RT-PCR method was used to detect the expression of PKAmRNA,CREBmRNA and BDNFm RNA in hippocampus of rats.Compared with the normal group,the contents of PKAm RNA,CREBm RNA and BDNFm RNA in the model group had no significant difference,but had a decreasing trend;Compared with the model group,the contents of PKAm RNA,CREBm RNA and BDNFm RNA in the high-dose group of Qiangzhifang,the low-dose group of Qiangzhifang,the Chaihu Shugan san group and the fluoxetine group also showed an increasing trend.Conclusion: 1.Chronic mild stress(CMS)can make ideal depression rat model.After modeling stimulation.Depression-like behavior of depressive rats such as Such as lack of interest and pleasure,decreased exploration ability,reduced activities can better simulate the depressive state of clinical depression patients.2.Qiang Zhi Fang can obviously improve the depression-like state of depression model rats.It is reasonable to explain the pathogenesis of depression from the standpoint of “yang yu shen tui”.3.Qiang Zhi Fang,which is based on the pathogenesis of “yang yu shen tui”.can play an antidepressant role by up-regulating the related genes and protein expression of c AMP-PKA-CREB signaling pathway. |
