Objective:To investigate the relationship between RNA binding protein HuR and the telomerase activity in oral tongue squamous carcinoma(TSCC).Methods:1.The tongue squamous carcinoma tissues and para-carcinoma tissues collected were detected separately telomerase activity with Telomeric repeat amplification protocol(TRAP)and the expression of HuR with Western blot.2.To test whether HuR can bind to TERC,the core component of telomerase,with RNA-pulldown and RNA-immunoprecipitation methods.3.To test whether HuR knockdown can regulate TERC or TERT,two core component of telomerase,with Western blot and RT-PCR.4.To test whether HuR knockdown can impact the combination of TERT and TERC while to test the effect of HuR knockdown on telomerase in SCC25 cells with TRAP method.5.Furthermore,to observe the telomere length with Southern blot while knockdown of HuR for a long time.Results:1.The telomerase activity and HuR expression level is significantly higher in tongue squamous carcinoma than that of para-carcinoma tissues(P<0.05).2.The results of RNA-pulldown and RNA-IP suggest that HuR can bind to the TERC.3.Knockdown of HuR not influences the expression level of TERC and TERT,but the assembly of TERC and TERT.Knockdown of HuR negatively regulates the telomerase activity.4.The telomere length becomes obviously shorter when knockdown of HuR for a long term.Conclusion:HuR could positively regulate telomerase activity and telomere length through binding to the TERC,which enhancing the assembly of telomerase in TSCC.
