Objective: Percutaneous coronary intervention(PCI)is the main method to treat coronary atherosclerotic heart disease(CHD).However,the occurrence of in-stent restenosis after operation has greatly affected its long-term efficacy.This experiment analyzed the difference of relative expression level of SENCR in peripheral blood between patients with in-stent restenosis(ISR group)and patients without restenosis(N-ISR group)after PCI,evaluated the diagnostic value of smooth muscle and endothelial cell-enriched migration/differentiation-associated long noncoding RNA(SENCR)for predicting the occurrence of ISR,and found new molecular markers for predicting the risk of ISR after PCI.Methods: To collect 70 cases of coronary heart disease(CHD)patients who were hospitalized in the Department of Cardiology,Affiliated Hospital of Jining Medical College from March 2017 to October 2018.They were treated with Percutaneous coronary intervention(PCI)for the first time and followed up successfully for 6-12 months.According to the results of coronary angiography(CAG),they were divided into in-stent restenosis group(ISR group,n=24)and in-stent non-restenosis group(N-ISR,n=46).The peripheral blood of the patients was collected.Firstly,the patients' monocytes were extracted with monocyte separating solution.And the total RNA was extracted with Trizol reagent.Finally,the relative expression level of SENCR in the twogroups was detected by real-time fluorescence quantitative PCR(RT-qPCR),and the difference was evaluated.Logistic regression analysis was used to explore the risk factors and independent risk factors of ISR.The ROC curve was used to evaluate the predictive value of SENCR expression in peripheral blood for the occurrence of ISR after PCI.Results:1.The relative expression level of SENCR in peripheral blood of ISR group after PCI was lower than that of N-ISR group(0.39±0.52,1.15±1.26),and the difference was statistically significant.(t=-4.11,P <0.05).2.Logistic regression analysis: Univariate logistic regression analysis of risk factors of ISR showed that there were significant differences in history of diabetes,smoking and SENCR between ISR group and N-ISR group(P<0.05).Logistic multivariate analysis showed that diabetes history and SENCR were independent risk factors for ISR.3.The ROC curve was used to calculate the area under the curve(AUC)to evaluate the predictive value of plasma SENCR relative expression in predicting the occurrence of ISR after PCI.The results showed that the AUC of plasma SENCR was 0.785,95% CI(Confidence Interval)is0.627 to 0.943.And there was statistical significance(P < 0.05).Conclusion:1.The occurrence of ISR may be related to the expression level ofSENCR,and the expression level of SENCR in ISR group was lower than that in N-ISR group.2.Low expression of SENCR is an independent risk factor for ISR.SENCR may be a new molecular marker for predicting the occurrence of ISR after PCI.
