Ganoderma atrum is one of the important herbal medicines in traditional Chinese pharmacy,with a variety of pharmacological effects.So far,a large amount of researches have been put on the functional ingredients and health effects of Ganoderma atrum.Among the varieties components,steroids poccessed a relatively high content in the fruit body and spores of Ganoderma atrum,and play various regulatory effects on the body.The main results are as follows:1.We compared the difference of the single factor experiment results with the indicators of ergosterol and free sterol,and finally design the response surface methology with free sterol as index.The parameters of the highest sterol content of Ganoderma atrum were as follows: 100 %,76.3 °C,4 h,the theoretical maximum yield of 6253 ?g / g,the test was verified to obtain the free sterol yield 6092 ?g/g.2.The antibacterial activity difference between ergosterol,ergosterol acetate and Ganoderma atrum sterol component were studied.In the inhibition test of bacteria,the antibacterial ability of Ganoderma atrum sterol component was higher than that of ergosterol and ergosterol acetate standards,but the three test samples showed no inhibition on both Penicillium.In addition,the separation and purification of sterol monomer compounds from the chloroform extract of Ganoderma atrum were separated and purified by Daigesol,?Bondapark and Kromasil columns to obtain the monomeric compound ergosta-7,22-dien-3-one.3.We also studied the antioxidant experiments of Ganoderma atrum sterol components by DPPH,MCC and ABTS scavenging assays.In in vitro antioxidant experiments,the Ganoderma atrum sterol component showed a certain antioxidant activity,and the highest free radical scavenging rate in several experiments was 25.86 ± 1.14%.In the study of the protective effect of hydrogen peroxide induced Caco-2 oxidative damage model,the addition of Ganoderma atrum sterol component and ergosterol inhibited intracellular free radicals and promoted the secretion of SOD and GSH-Px to a certain extent.It inhibits the production of MDA and the release of LDH,thereby protecting oxidatively damaged cells and increasing cell viability.In addition to the optimization of the traditional leaching process and the separation and purification of the monomeric compounds,and the sterol component of Ganoderma atrum was used as in this research to study the biological activities,aimed to provide theoretical basis for health food and drug development to some extent.
