Objective: To synthesize a lactose-targeted acaldodextran nano-drug carring doxorubicin and evaluate its efficacy on liver cancer cell lins.Method: The experiment was divided into four parts: 1 The nanometer materials were synthesized by the acetal reaction of dextran,Then,they were covalent binded with doxorubicin by dissolving dialysis method,at last,the acaldodextran macromolecule nano-drug delivery system carring doxorubicin(AcDex@Dox)were prepared.Lactose was grafted onto the acaldodextrans' surface,and then,they were synthesized by lactose-acaldodextran(Lac-AcDex)in an organic solvent,using the amino covalent bond of doxorubincin conjugated with them to form the lactose-targeted acaldodextran macromolecule nano-drug carring doxorubincin(Lac-AcDex@Dox)at last.2 The diameters and surface features of these nanoparticles were examined by transmission electron microscopy and dynamic light scattering,and the functional group were characterized by infrared spectrum and nuclear magnetic resonance hydrogen spectrum,and the release quantity of nanodrugs were determined by in vitro drug-release test under different PH.3 The nanoparticle drug entering cells' nucleus exhibited an obvious cytotoxic effect on HepG2 cells by in vitro cell apoptosis experiments including MTT assay and drugs swallow reaction.Lastly.4 we compare the difference of small molecules adriamycin and macromolecule nano-drugs by animor tumor suppression,Meanwhile,we analysis biodistribution of the Kunming mice which were injected with nano-drugs and the control group,to test the biocompatibility of nanoparticles.Results: 1.Lac-Acdex@Dox was well-dispersed in water,homogeneous and stabilized.2.The doxorubicin nanoparticle has more drug release under weak acid conditions(P<0.05).3.Nano-drugs can be effectively taken in by HepG2 cells through endocytosis process,and finally enter the nucleus,resulting strong cytotoxicity effect.4.Lac-Acdex@Dox has the best tumor inhibition effect compared with the contrlo group(saline,smll molecule adriamycin and Acdex@Dox)(P<0.01).Conclusion: 1.We have successfully synthesized a lactose-targeted acaldodextran nano-drug delivery system containing doxorubicin,and the drug delivery system is well in stability.2.The doxorubicin nano-drug has better tumor inhibition effect than the small molecule doxorubicin,and smaller toxic side.
