Objective:Discussion the influence of Curcumin nicotinic(CurTn)for balloon aortic vascular smooth muscle cell proliferation after injury caused by rat's abdomen and its possible mechanism.Methods:50 male SD rats,randomly divided into sham operation group(SOG)?curcumin group(CG)?low-dose CurTn group(LCTG)?high-dose CurTn group(HCTG)?model group(MG),each group of 10.The SOG and the MG were filled with equal volume of physiological saline into the stomach every day.CG,LCTG and HCTG were respectively given daily CurTn 100mg/Kg,CurTn 10mg/Kg,and CurTn 100mg/Kg for gastric intervention.The left carotid artery ligation was performed in the sham operation group on the third day of the gastric lavage.CG,LCTG,HCTG and MG were injured by the abdominal aortic balloon after the third day of continuous gastric lavage.It was found that that abdominal aorta sample was taken after 14 day after postoperative continuous drug filling,and perform the following tests:1?The diameter of the tube cavity ?the thickness and the ratio of intima and tunica media was observed by HE staining.2?Fluorescence staining observed Daxx localization and the number of?-smooth muscle actin(?-SMA).3?Western Blot method was used to detect the expression of Daxx,STAT3,PTNE and p-akt in vascular tissues.Result: 1.HE staining display:The vascular integrity of the SOG was good,and no neointimal formation was observed.Intimal thicken of that MG was apparent,and the constriction of lumen was significantly higher than in the other group;Compared with CG and LCTG,the ratio of intima and tunica media thickness in the HCTG was lower than that in the other two groups(P< 0.05).2.Fluorescence staining shows:The expression of Daxx/ ?-SMA in the sham group was stable.However,there was a significant increase in Daxx/ ?-SMA expression in the other groups.3.Western Blot shows:The expression of related proteins detected by each group of balloon injury was higher than that in the sham group(P< 0.05).The difference was statistically significant.The expression of Daxx/PTEN was significantly increased in the HCTG compared with other balloon injury patients(p < 0.05).And the protein expression of STAT3/ p-akt was significantly lower than that of other balloon injuries(p < 0.05).The above data was of statistical significance.Conclusion: 1?CurTn can inhibit the proliferation of vascular smooth muscle cells after balloon injury in rat abdominal aorta;2?CurTn inhibits the proliferation of rat vascular smooth muscle cells may be related to upregulation of Daxx/STAT3/PTEN signaling pathway.
