Objective:Gastric cancer?GC?ranks 5th in the world in terms of morbidity and 3rd in terms of mortality.China is a region with a high incidence of gastric cancer,which poses a great threat to the life and health of Chinese people.To explore the expression level of hsa circ 0058132 in gastric adenocarcinoma?GA?tissues and analyze the relationship between it and the clinicopathological features of GA patients.To investigate the effect of changing in expression level of hsa circ 0058132 on the biological behavior of GA cells.Aim to reveal the pathogenesis of GC at the molecular level,and to search for a new biomarkers for GC's detection and a new targeted treatment point.Methods:?1?Gene chips of high flux assay were used to detect the expression profiling of circRNA in GA tissues,screening out a target circRNA were not reported in the literature before.?2?Collecting tissue samples,then use qRT-PCR to detect the expression level of hsa circ 0058132 in GA,and analyze the relationship between it and the clinicopathological features of GA patients.?3?siRNA interference was used to inhibit the expression level of hsacirc0058132 in GA cells,verifing the effect of hsacirc0058132 on proliferation,migration,invasion,apoptosis and cell cycle of GA cells by cell function experiments.Results:?1?The expression of hsacirc0058132 in GA was up-regulated by 60.2%?P<0.05?,its up-regulated fold change 9.71 compared with the paracancer tissue.?2?The expression of hsacirc0058132 was statistically significantly associated with patient's TNM staging,lymph node metastasis of GA?P<0.05?.?3?hsa circ 0058132 was significantly up-regulated in 5 GA cell lines?P<0.05?,inhibiting the expression of hsacirc0058132 can reduce the proliferation,migration and invasion ability of GA cells?P<0.05?,as well as affect the distribution of cell cycle,and promote the apoptosis of GA cells?P<0.05?.Conclusions:hsacirc0058132 was upregulated in GA,the high expression is closely related to TNM staging and lymph node metastasis in patients with GA;Inhibiting the expression of hsacirc0058132 can affect the biological behavior of gastric adenocarcinoma cells.It can be used as a potential biomarker for GA's screening and prognosis.
