Study On Combined Administration System Of Piperine And Curcumin Gastric Floating Microspheres

Nowadays,the studies on the combination of curcumin and piperine are mainly focused on solid dispersion,cyclodextrin inclusion compound and compound micro-emulsion.These single dosage forms have been shown to improve the in vitro stability of curcumin in the formulation,but the in vivo stability and pharmacodynamic effects have to be investigated further.In particular,the physical and chemical properties of the two drugs were not taken into account with regard to the directed administration of the drugs.In order to reduce the characteristics of the enzyme on the degradation of curcumin,the potential value of the combined administration of the two drugs can be obtained in this study by contacting with piperine to compete with biological enzymes in the organism.Finally,the hollow slow release microspheres and piperinecyclodextrin inclusion compound were combined in a certain form to increase the solubility,dissolution rate and bioavailability of the two natural drugs,so as to improve the efficacy of the combination.Improving the utilization pattern of traditional Chinese medicine resources provides new motivation for preparing new dosage forms of traditional Chinese medicine and improving the competitiveness of traditional Chinese medicine in the pharmaceutical market.The main work contents are as follows:1.Briefly introduce the current situation and trend of natural drug research at home and abroad,the problems existing in Chinese natural drug research,the pharmacological properties of curcumin,the pharmacological properties of piperine,the advantages and research progress of the combination of curcumin and piperine.2.Curcumin is prepared into gastric floating microspheres by using a new technique of gastric flotation microspheres.Curcumin can stay in the stomach for a long time and release slowly.It not only increases the drug absorption,improves the bioavailability,but also avoids the peak-to-valley effect,but also improves the local efficacy,facilitates the administration of drugs to patients and improves the compliance.3.The inclusion experiments of target molecule piperine with chemically modified ?-cyclodextrin(ethylenediamine-?-cyclodextrin) were carried out to investigate the inclusion effect of ethylenediamine-?-cyclodextrin on piperine molecules and the release degree of the drug in vitro.The specific inclusion effect of ethylenediamine-?-cyclodextrin on piperine in aqueous phase was verified,which could effectively improve the stability and solubility of piperine,and lay a foundation for the study of the combination of curcumin gastric floating microspheres and piperine/ cyclodextrin inclusion compound.4.To simulate the inclusion mode of cyclodextrin derivative and piperine under the help of molecular simulation software,the theoretical inclusion mode,combined with the experimental results of inclusion complex formed by cyclodextrin derivative and piperine has enriched the research results from both theoretical and experimental angles.5.The feasibility of constructing the combination delivery system of curcumin/ piperine will be explored,combined with the latest literature reports at home and abroad,and the newly developed latest administration technology and dosage form will be taken as reference to provide reference and research ideas for the development of the combination delivery system of curcumin gastric floating microspheres and piperine/cyclodextrin inclusion compound.Finally,the hollow slow release microspheres and piperine-cyclodextrin inclusion compound were combined in a certain form to increase the solubility,dissolution rate and bioavailability of the two natural drugs,so as to improve the efficacy of the combination.Improving the utilization pattern of traditional Chinese medicine resources and provides new motivation for preparing new dosage forms of traditional Chinese medicine and improving the competitiveness of traditional Chinese medicine in the pharmaceutical market.
