Purpose:To explore the possible mechanism of mir-193a-3p affecting adriamycin resistance in McF-7 /ADR cells,so as to provide experimental basis for finding new chemotherapy targets and reversing adriamycin resistance.Methods:(1)By Lipofectamine ?(liposome)2000 miR-193-a-3 p analog(mimic)and inhibitor(inhibitor)respectively transfection into MCF-7 and MCF-7 / ADR cells,respectively,set of micrornas segments of negative group(negative control)and blank control(2)cell proliferation and proliferation inhibition under different concentrations of drugs were detected by Thiazolyl Blue Tetrazolium Bromide(MTT)after cell transfection.(3)bioinformatics website was used to predict the downstream regulatory protein of mir-193a-3p,Recombinant Cyclin D1(CCND1),and gene function analysis was conducted.(4)the use of real-time fluorescent quantitative polymerase chain reaction lock(policy real-time polymerase chain reaction,qRT-PCR)to detect the contents of transfection efficiency,messenger RNA.(5)the differential expression of CCND1 protein in cells was detected by Western Blot(WB).Results:(1)The expressions of mir-193a-3p and CCND1 in McF-7 and McF-7/ADR cells were different(2)The transfection of mir-193a-3p mimics,inhibitors and si-ccnd1 was verified by PCR(3)Drug resistance of McF-7/ADR cells decreased after overexpression of mir-193a-3p(4)McF-7/ADR resistance to adriamycin decreased after CCND1 silencing(5)Overexpression of mir-193a-3p had no significant effect on the migration ability of McF-7 /ADR cells(6)The bioinformatics website predicted that the downstream target gene of mir-193a-3p might be CCND1 protein(7)Overexpression of mir-193a-3p can down-regulate the expression of CCND1 proteinConclusion: Mir-193a-3p may reverse adriamycin resistance in breast cancer McF-7 /ADR cells by inhibiting the expression of CCND1...
