Gastric cancer as a malignant tumor of digestive tract has been a serious threat to the national health.At the present stage,the level of drug treatment for gastric cancer has made great progress but there are still difficult problems such as severe toxic and side effects,high recurrence rate and high mortality.As a common folk medicine widely distributed in China,Panax japonicus.C.A.Mey is one of the characteristic medicinal plant resources in hubei province.A large number of literatures have proved that Panax japonicus.C.A.Mey has anti-fatigue,anti-oxidation,anti-inflammatory,gastrointestinal protection and other effects,the main active constituents responsible for these pharmacological effects are Chikusetsusaponins.However,the research foundation on the anti-cancer effect of Chikusetsusaponins monomers is still relatively weak.Objective:Based on this,this study started from the cell level to study the effect of Oleanane-Type triterpene saponins of Panax japonicus.C.A.Mey on the proliferation,migration,invasion and apoptosis of human gastric cancer cells SGC-7901,so as to provide a new drug source for the treatment of gastric cancer.Methods:The experiment was divided into three groups:a blank control group,a positive control group(5-fluorouracil)and a drug group(Chikusetsusaponins).Different concentrations of Chikusetsusaponin ?,?a and ?were respectively applied to SGC-7901 cells in vitro.The morphological changes were observed by microscope after 24h.The inhibitory effect of Chikusetsusaponins on the proliferation of gastric cancer cells was assessed by CCK-8 assay.The apoptosis of gastric cancer cells was analyzed by Flow cytometry.The effects of Chikusetsusaponin on the migration and invasion of gastric cancer cells were detected by scarification test and transwell migration assays.Results:Compared with the blank group,the morphology of cells in the drug group was significantly changed,with obvious cell edge shrinkage and irregular cell morphology.Chikusetsusaponin ?,?a and ? showed a significant concentration-dependent proliferation inhibition in gastric cancer cells in the range of 37.5 to 600 ?g/mL after 48h of treatment,especially the Chikusetsusaponin IVa.The Flow cytometry results showed that Chikusetsusaponin ?,?a and ? induced the apoptosis of gastric cancer cells at the different degree at 250 ?g/mL and 500 ?g/mL after 48h treatment,especially the Chikusetsusaponin ?.And,Chikusetsusaponin ?,?a and ? can significantly inhibit the migration and invasion of gastric cancer cells at 250 ?g/mL and 500 ?g/mL for 24h.Conclusion:Oleanane-Type triterpene saponins of Panax japonicus.C.A.Mey could significantly induce the apoptosis of gastric cancer SGC-7901 cells and inhibit their proliferation,invasion and apoptosis.
