| ObjectiveTo clarify the effect of inhibiting the activity of stearoyl-CoA desaturase 1(SCD1)on the proliferation of breast cancer MCF-7 cells and to initially explore its mechanism.Methods1.The cultured human breast cancer MCF-7 cells were divided into SCD1 inhibitor A939572 group,SCD1 inhibitor CAY10566 group and negative control group(NC group).2.The proliferation ability of human breast cancer MCF-7 cells was detected by MTS and EdU staining.The proliferation of single breast cancer MCF-7 cells was detected by cell clone formation.3.The expression of proliferation-related genes in breast cancer MCF-7 cells treated with SCD1 inhibitors were detected by qRT-PCR.The mechanism of inhibition of SCD1 on proliferation of human breast cancer MCF-7 cells was investigated and analyzed.ResultsMTS experiment showed that the proliferation of human breast cancer MCF7 cells was decreased after treatments SCD1 of inhibitor(P<0.05).EdU experiment showed that the proliferation of h uman breast cancer MCF7 cells was decreased after the addition of SCD1 inhibitor(P<0.05).Cell clone formation experiments showed that the proliferation of human breast cancer MCF7 cells was decreased after the addition of SCD1 inhibitor(P<0.001).qRT-PCR experiments showed that the transcription of the relevant proliferation genes cyclin-D1,CDC6,ATF3,EMP1,and ACVR1 was significantly inhibited after the addition of SCD1 inhibitor(P<0.001).Conclusions1.Inhibition of SCD1 activity inhibits breast cancer cell proliferation2.Inhibition of SCD1 activity may inhibit proliferation of human breast cancer MCF-7 cells by reducing the transcription levels of the proliferation-related genes CyclinD1,CDC6,ATF3,EMP1,ACVR1. |
