BackgroundHyperlipidemia is a common frequently-occurring disease and an important risk factor for cardiovascular and cerebrovascular diseases such as coronary heart disease and stroke,a survey in 31 provinces(autonomous regions and cities)in China in 2012 showed a significant increase in the prevalence of hyperlipidemia.And there is a serious trend of rejuvenation,so slowing down the incidence of hyperlipidemia and the prevention and treatment of secondary diseases has become an urgent problem to be solved in the medical community.With the development of society,people's demand for treatment methods is more and more inclined to have both curative effect and green non-toxic side effects,moxibustion is one of the characteristic therapies of traditional Chinese medicine,and its function is quite many."Materia Medica from new" said:"Ai Ye Kuxin,warm and ripe heat," Pure yang nature,can return to the absolute yang,through the twelve meridians,walk the three yin,regulate qi and blood,cold and dampness,warm the uterus,.In order to moxibustion fire,can penetrate the meridians and eliminate 100 diseases,"at present,many clinical studies have confirmed that moxibustion has definite curative effect,safety,no toxic and side effects,easy operation and no pain,and has been paid more and more attention and favored by patients.In view of the fact that moxibustion has a definite clinical effect on lipid regulation,relevant experimental studies have been carried out in this study.Through literature review and clinical observation on the etiology and pathogenesis of hyperlipidemia and moxibustion,it was found that the main etiology and pathogenesis of hyperlipidemia were yang deficiency and qi stagnation,blood stasis and phlegm obstruction.Moxibustion has the characteristics of warming yang qi,regulating qi,warming phlegm and blood stasis,warming and dredging blood vessels,and can be well targeted to treat hyperlipidemia,so moxibustion is selected as the method of regulating hyperlipidemia.Then,based on the ancient and modern physicians' understanding of the related factors affecting moxibustion effect(moxibustion feeling and moxibustion quantity),as well as the inventory of relevant modern research,there are a lot of studies on moxibustion sensation(especially different moxibustion temperatures).However,there are few studies related to moxibustion volume,and most of the existing studies focus on the comparison of different moxibustion time,and there is little research on the amount of burning wormwood per unit time(that is,different moxibustion area and burning speed per unit time).Then the subject also looks for the relevant theoretical basis from the classical literature,such as Sun Simiao's "preparing for a thousand gold prescriptions",which emphasizes that the disease should be in the superficial part of the body,the amount of moxibustion should be small,and the amount of moxibustion should be large in the internal part of the disease,and that the disease should be in the superficial part of the body,and the amount of moxibustion should be large in the internal part.And "head face pharynx,moxibustion most want to give birth to less.”Chest,back and abdomen moxibustion,especially ripe.Within the abdominal cavity,in order to be greedy for sex in the five flavors,never tired of becoming a disease,wind-cold knot,water and grain,should be ripe,"as well as" acupuncture and moxibustion birth meridian moxa size "also said:" such as abdominal diseases,addictive bumps,such as Fu Liang Qi,and so on,but must be big moxa cone." "knowing that chronic metabolic diseases such as hyperlipidemia are in the internal organs,it is advisable to use large amounts of moxibustion and moxibustion in the abdomen,and because the abdomen follows the three meridians of the liver,spleen and kidney,and where the Ren pulse is located,Liver,spleen and kidney are the main related viscera of hyperlipidemia,which provides a certain theoretical basis for the treatment of hyperlipidemia with large amount of moxibustion on the whole abdomen in this experiment,in many experimental studies of moxibustion and lipid regulation,Most of the selected acupoints were Shenque acupoint.Therefore,different moxibustion quantities(that is,simple Shenque acupoint moxibustion and large moxibustion volume moxibustion in the whole abdomen)were taken as the different intervention factors in this study.Finally,for the selection of observation indexes,this study through the comprehensive review of moxibustion lipid-regulating related experimental research,It is known that the current research on the mechanism of moxibustion and lipid regulation and its prevention and treatment of AS in addition to exploring the related proteins regulating lipid metabolism(observing the contents of serum apoA,apoB and lipoprotein metabolic enzymes(LPL and HL activities),protecting vascular endothelial structure(electricity).Microscopic observation of the ultrastructure of aortic endothelial cells,protection of vascular endothelial function(observation of serum ET-1,NO content),antioxidant effect(observation of serum OX-LDL,LPO,SOD.GSH?px content),anti-inflammatory effect(observation of serum.The contents of IL-6,IL-8,TNF-a,etc.)and the regulation of the expression of related receptors(observing the expression of CGRP.the expression of vanillic acid receptor ImRNA in dorsal root ganglion transient receptor potential,and the expression of gap junction protein 43 in the skin at moxibustion).There is very little research on other aspects;In the literature review,it is also known that apoA-l/ABCAl pathway promotes cholesterol reverse transport one of the important ways in(RCT),when the pathway is developed,the excess cholesterol efflux from cells is carried by apoA-1 in HDL and transported to the liver for degradation.Although the existing research has well expounded the possible mechanism of moxibustion regulating lipid and protecting blood vessels to a certain extent,it is not very comprehensive,so this experiment starts with different moxibustion quantities to observe and explore the role of moxibustion in lipid regulation.And from the apoA-l/ABCA1 cholesterol reverse transport pathway to explore the possible mechanism of moxibustion to protect blood vessels.ObjectiveTo observe and compare the effects of different moxibustion doses on blood lipid level,endothelial function and apoA-1/ABCA1 cholesterol reverse transport pathway in hyperlipidemic mice.To explore the possible mechanism of moxibustion on regulating lipid and preventing and treating atherosclerotic(AS).MethodsForty 2-month-old male C57BL6 mice of SPF grade were selected and fed adaptively for 7 days.They were randomly divided into blank normal control group (group CON),hyperlipidemia model group(group MOD)?Shenque acupoint moxibustion group(group Tl)and abdominal moxibustion group(group T2)with 10 mice in each group,the body weight of each group was measured and recorded before the model was established.In the whole process of modeling and experiment,except for the normal diet of group CON,the other three groups were fed with high-fat diet, after 30 days of feeding,the following interventions were carried out:group Tl fixed mice,Moxibustion with aged wormwood thread incense(diameter 2mm)was suspended directly above Shenque acupoint,the distance from the skin was about 4-6 mm.and the ash was bounced every 5 seconds.In group T2.the diameter 20mm of Artemisia argyi was suspended vertically at(20±5)mm above the whole abdomen.and the ash was bounced every 5 seconds.During moxibustion in group T1 and group T2,the temperature of Shenque acupoint skin area was monitored by contact thermometer every other 1 min,and the moxibustion temperature was controlled at 43?-45? by adjusting the distance between moxibustion and skin up and down,10 min each time,once a day for 30 days.During this period,the diet is still high in fat.group CON and group MOD:the mice in the two groups were fed with basic diet and high-fat diet respectively,which were routinely fixed for 10 minutes every day,and the others were not treated once a day.after 30 days of intervention,the body weight of mice in each group was measured and recorded,and the serum and fresh thoracic aorta of mice were taken.Serum TG,TC,HDL-C,LDL-C,apoA-1,ET-1,NO and thoracic aorta western Blot were detected for ABCA-1 protein expression.Results1.There was no significant difference in body weight between each group before modeling(P>0.05),but the body weight in group MOD was significantly higher than that in group CON after intervention(P<0.05).Compared with group MOD,the body weight of group T1 and group T2 decreased significantly(P<0.05),especially in group T2(P<0.05).2.the contents of serum TG,TC,LDL-C and HDL-C in group MOD were significantly higher than those in group CON(P<0.05),group T1 and group T2,but the contents of serum TG,TC and LDL-C in group T1 were lower than those in group MOD.However,the levels of serum TG,TC and HDL-C were significantly decreased in the group with no significant difference(P>0.05):group T2,and the content of LDL-C was improved,but the difference was not significant(P>0.05).3.compared with group CON,the level of apoA-1 in group MOD was significantly higher than that in,group MOD after high-fat diet(P<0.05).The levels of serum apoA-1 in group T1 and group T2 were lower than those in group MOD,but there was no significant difference between group T1 and group T2 and group CON(P>0.05);group T1.The content of serum apoA-1 was significantly higher than that of group CON.compared with group CON,the content of serum apoA-1 in group T2 was slightly higher than that in group CON,but the difference was not significant(P>0.05).4.the content of serum nitric oxide(NO)in group MOD was slightly higher than that in group CON,but the difference was not significant(P>0.05).Compared with group MOD,the content of NO in group T1 and group T2 increased in turn,but there was no significant difference between the two groups(P>0.05).The content of serum endothelin-1(ET-1)in group MOD was significantly higher than that in group CON(P<0.05).The content of ET-1 in group T1 and group T2was significantly lower than that in group MOD(P<0.05),especially in group T2(P<0.05),and it was the most significant in group T2(P<0.05),especially in group T2(P<0.05).5.The gray value of the strip image detected by western Blot was scanned by software,the statistical results showed that the expression level of ABC A1 in thoracic aorta of mice in group MOD was significantly lower than that in group CON(P<0.05).After different intervention,group T1 and group T2 was compared with group MOD,The expression of ABCA1 was significantly increased in both groups,especially in group T2.Conclusions1.A high-fat diet can lead to rapid weight gain,Moxibustion therapy can slow down the rate of weight gain;2.moxibustion therapy can slow down the rate of weight gain.2.moxibustion therapy can slow down the rate of weight gain.Moxibustion with large amount of moxibustion on the whole abdomen has better effect on regulating lipid,promoting reverse transport of cholesterol and protecting vascular endothelial function than simple moxibustion at Shenque point.3.Moxibustion for lipid regulation may be related to the apoA-1/ABCA1 pathway mediating cholesterol reverse transport.
