ObjectiveBy analyzing the data of physical examination of the population in the army’s cadre rest centre in Beijing,the normal reference ranges of T and B lymphocyte subsets and cytokines in different age groups of the population were compared and preliminary established.And the effects of Thymic peptide analogues and Cordyceps sinensis powder analogues on the lymphocyte subsets and cytokines in human body were explored.Methods1、A total of 724 healthy people were selected from 868 people who participated in the physical examination in Department of Geriatrics,General Hospital of PLA in 2017 and 2018.They were divided into four groups according to age below 60,60-69,70-79 and over 80.The percentage of lymphocyte and subsets of lymphocyte in peripheral blood were detected by two-color flow cytometry combined with hematology counter,and cytokines were detected by suspension chip system.SPSS22.0 software was used to compare the statistical differences of various indicators among different groups,and the normal reference range of lymphocyte subsets and cytokines for the elderly in Beijing was obtained by using the results of the screened population aged over 60 years old.2、Among the 868 people who participated in the examination,114 people who used immunomodulators were selected and divided into three groups according to different kinds of immunomodulators.People who did not use immunomodulators were divided into the control group.The lymphocyte subsets and cytokine counts of the these groups were compared among groups respectively to find statistical differences.Results1、By analyzing the percentage of lymphocyte subsets and cytokine counts in p eripheral blood,it is concluded that the percentage of lymphocyte decreases with ag e.In the percentage of lymph subsets,CD3~+,CD3 CD4~+,CD4 45RA~+,CD8~+CD28~+,CD4~+ HLADR~-in the elderly over 80 years old were significantly higher than those under 60 years old,while CD28~+,CD3~+ HLADR~-were significantly higher than tho se under 70 years old.Cytokine counting:IL-17 decreases with age;IP-10 increase s with age;IL-4 and IL-9 in the elderly over 60 years old are lower than those un der 60 years old,while IL-1β is higher than those under 60 years old;PDGF-BB a nd TNF-a in the elderly over 70 years old are lower than those under 70 years ol d,and IL-8 is lower than those under 60 years old.2、Statistics showed that the normal reference range for the percentage of peri pheral blood lymphocyte subsets of the elderly in Beijing area was as follows(expr essed in between the groups using immunomodulators and the control group,it was found that:in the distribution of lymphoid subsets,the CD3~+,CD28~+,CD8~+CD28~+,CD3~+CD4~+,CD4~+45RA~+,CD3~+HLADR~-measured by the group using Thymic peptide analogues decreased compared with the control group,and the CD3~+HLADU-measured by the group using Cordyceps sinensis powder analogues and the combination of Thymic peptide analogues and Cordyceps sinensis powder analogues decreased compared with the control group.In terms of cytokine count,the levels of IL-1β,IL-4 and IL-6 in the Thymic peptide analogues group were higher than those in the control group,while the levels of IL-10 and TNF-α in the immunomodulator group were lower than those in the control group.Comparisons between groups using different immunomodulators were found:In terms of lymphocyte subsets distribution,CD3~+CD4~+,CD8~+CD28~+,CD45RA~+,CD4~+45RA~+ measured by Thymic peptide analogues alone group were lower than those by Cordyceps sinensis powder analogues alone group,while CD45RO~+was higher than that by Cordyceps sinensis powder analogues alone group;in terms of cytokine counts,IL-1β,IL-4 and IL-6 in the combination of two analogues and Thymic peptide alone groups were higher than those in the Cordyceps sinensis powder analogues group,while TNF-α was lower than those in the Cordyceps sinensis powder analogues group.ConclusionsWe found the distribution of some lymphocyte subsets CD3~+,CD3~+CD4~+,CD4~+45RA~+,CD8~+CD28~+,CD4~+HLADR~-,CD28~+,CD3~+HLADR~-show increasing trends with the increase of age,and the counting of cytokines IL-4、IL-8、IL-9、IL-17、PDGF-BB、TNF-α show decreasing trends with the increase of age,suggesting that the risk of infection,imnune system diseases and tumors increased in this age group.The normal range of lymphocyte subsets and cytokines suitable for the elderly was preliminarily summarized for clinical reference.At the same time,the specific targets of Thymic peptide analogues and Cordyceps sinensis powder analogues on lymphocyte subsets and cytokines were found through the analysis of the object using immunomodulators in the study subjects.Thymic peptide analogues mainly affect the distribution of CD3~+,CD28~+,CD8~+CD28~+,CD3~+CD4~+,CD4~+45RA~+,CD3~+HLADR and the cytokines IL-1β,IL-4,IL-6,IL-10 and TNF-α,while Cordyceps sinensis powder analogues only affect IL-10.
