| Objective:Firstly,the expression of CXCL17 in gastric cancer and adjacent normal gastric tissues was analyzed.The relationship between differential expression and prognosis of gastric cancer patients was analyzed.Then the relationship between CXCL17 and gastric cancer cell proliferation in vitro was studied to illustrate the relationship between CXCL17 expression and gastric cancer.Methods:1.Under the R language environment,grab the data of CXCL17 and gastric cancer in TCGA and GEO database,analyze the expression of CXCL17 in gastric cancer tissues and adjacent normal gastric tissues,and the relationship between the expression of CXCL17 in gastric cancer and the 10-year disease-free survival rate.2.The pathological tissues of 90 cases of gastric cancer were collected and the expression of CXCL17 in gastric cancer tissues and corresponding normal gastric tissues was detected by tissue microarray immunohistochemistry.The survival time(months)of the above 90 patients was followed up.Kaplan-Meier survival curve was used to analyze the relationship between CXCL17 expression and prognosis in gastric cancer.3.MTT assay was used to observe the proliferation rate of gastric cancer cells in MGC803 and SGC7901 cell lines treated with 50,100,200,400ng/ml CXCL17 medium for 24h,40h,56h and 72h respectively,with was used to analyze the effect of CXCL17 on the proliferation of gastric cancer cells.Results:1.The results of GEO and TCGA data showed that the expression of CXCL17 in normal tissues was higher than that in normal gastric tissues(p<0.05).The effect of CXCL17 expression on survival rate was Kaplan-Meier survival curve,and patients with high expression of CXCLI7 had a significantly lower lo-year disease-free survival rate(p<0.01).2.Tissue microarray immunohistochemistry showed that the expression of CXCL17 in the nucleus was higher in gastric cancer than in adjacent normal tissues(p<0.01),However,there is no significant relationship between the current statistical analysis and age,gender,tumor location,tumor size,pathological stage,lymphatic metastasis,and depth of invasion(P>0.05).Kaplan-Meier survival curve analysis showed that the survival rate of the lower expression group of CXCL17 high expression group decreased(P<0.05).3.MTT results The proliferation rate of gastric cancer cells treated with CXCL17 concentration of 100 ng/ml culture medium increased significantly(p<0.05).Conclusions:1.The expression of CXCL17 in the primary gastric cancer is significantly higher than that in the normal gastric tissue.2.CXCL17 has a low survival rate in patients with gastric cancer with high expression in gastric cancer.It can be used as a potential indicator for predicting the prognosis of gastric cancer.3.CXCL17 promotes the proliferation of gastric cancer cells. |
