Objective:Using Caenorhabditis elegans as a model animal to study the damage of the reproductive system caused by ionizing radiation,and to explore the protective effect of polydatin on the radiation damage of the C.elegans reproductive system.Methods:1.Using 60Co y-rays to irradiate wild-type C..elegans with 50Gy,100Gy and 200Gy at one time,and observe the changes of reproductive ability and ovipositor structure of C.elegans after irradiation.2.AO fluorescence staining was used to measure the level of germ cell apoptosis.H2DCFDA fluorescent probe method was used to detect the level of ROS in vivo.The apoptosis regulatory genes cep-1 and egl-1 mRNA were detected by real-time fluorescent quantitative PCR(QRT-PCR).3.Using 50μM,100μM,200μM concentration of polydatin to intervene wild type nematodes,observe the changes of lifespan and reproductive ability of C.elegans,and screen the appropriate drug concentration.4.The appropriate concentration of polydatin and pretreatment of C.elegans were used to detect the changes of ROS and germ cell apoptosis in C.elegans,and the protective effect of polydatin on the reproductive system of C.elegans was evaluated.Results:1.When the dose of 60Co y-ray irradiation reached 50Gy,the reproductive system damage occurred.The reproductive ability of C.elegans decreased and the rate of genital malformation increased.As the dose of radiation increases,the damage to the reproductive system becomes more pronounced.2.6 Co y-rays can increase the level of ROS in the nematodes,and the number of germ cell apoptosis and the expression of apoptosis regulators cep-1 and egl-1 mRNA are significantly increased.3.100μM polydatin will not cause toxic effects on C.elegans,prolong life,and higher concentration will inhibit its reproductive ability.100μM polydatin is a reasonable drug intervention concentration.4.100μM polydatin can improve the reproductive ability of C..elegans after irradiation,reduce ROS levels in vivo,and reduce germ cell apoptosis.Conclusion:In this study,we found that 60Co γ-rays can induce the increase of ROS levels and germ cell apoptosis in C.elegans,causing damage to the reproductive system of C.elegans.Polydatin has the effect of reducing ROS levels in the C.elegans and inhibiting apoptosis of germ cells,and has a preventive effect on radiation-induced reproductive damage.
