In this thesis,UPLC-ESI-MS/MS method was used to qualitatively and quantitatively detect the curcuminoids in turmeric medicinal materials from different countries,and the content of main components in turmeric medicinal materials collected from different geographical regions was compared.The experimental results show that the turmeric of Thailand always contains the highest amount of curcuminoids.A DART-MS/MS method was established to rapidly determine the three main components in different turmeric herbs,and the measured results were compared with those determined by UPLC-ESI-MS/MS.The results show that the method not only fast but accurate.Curcumin has many biological activities,but its low water solubility and poor stability hinder its clinical development and application.While liver fibrosis is becoming more and more serious in China,to increase the water solubility of curcumin constituents by three kinds of food grade surfactants,and to study the effect of surfactants on the quality standard and stability and anti-hepatic fibrosis in vivo of curcumin solution.LC-MS/MS analysis showed that the structure of curcuminoids was stable;The results of fingerprinting analysis demonstrated that the similarity of 18 batches of samples were higher than 99%.The contents of curcumin,demethoxycurcumin and bisdemethoxycurcumin in different batches of samples were stable;the solubility of curcuminoids was increased 517 times compared with the free state(2.677 ?g/mL)after solubilization with surfactants,and repeated tests indicated that the structure and contents of curcuminoids were stable;the solubilized curcuminoids exhibited better anti-liver fibrosis effects than the free.In this paper,the in vivo metabolism of curcuminoids after oral administration was analyzed.The curcumin and Curcumin Glucuronide in rat plasma were characterized by UPLC-QTOF-MS/MS and UPLC-ESI-MS/MS.Quantitative analysis and pharmacokinetic analysis of the data using WinNonlin software.The results showed that the bioavailability of the curcuminoids after solubilization was higher,which provided a new idea for the clinical application of poorly soluble components.
