Objective:To systematically evaluate the effect of SRSF2 gene mutation on the prognosis of patients with myelodysplastic syndrome.Methods:Databases,including PubMed,Embase and the Cochrane Library,were searched for relevant studies published up to December 2018.All cohort studies were screened according to inclusion/exclution criteria.Data extraction and bias risk assessment were conducted for the eligible literature.The observed indicators were statistically processed by RevMan 5.3 software.Overall survival(OS)was selected as the primary endpoint,acute myeloid leukemia(AML)transformation and the progression free survival(PFS)were the secondary endpoint.We extracted the corresponding hazard ratios(HRs)and their 95% confidence intervals(CIs)for OS,AML transformation and PFS from multivariate Cox proportional hazardsext models.Results:A total of 11 cohort studies were included in the final meta-analysis.Our results indicated that SRSF2 mutations had an adverse prognostic impact on OS[HR=1.63,95%CI(1.36,1.94),P<0.00001],PFS[HR=6.20,95%CI(2.64,14.58),P<0.00001]and AML transformation[HR=2.02,95%CI(1.49,2.74),P<0.00001]in the total popution.Conclusion:This meta-analysis indicates an important,adverse prognostic impact of SRSF2 mutations on OS,PFS and AML transformation in patients with MDS.The identification of mutations in SRSF2 can improve risk stratification and help make treatment decisions.
