Objective:To observe the early pathological changes of renal tubular and peritubular capillary?PTC?in renal tissues of mice after administration of contrast medium.Methods:70 C57BL/6J mice were selected,the normal group N?n=35?and the model group CM?n=35?were set up.Mice in CM group were injected intraperitoneally with iopamidol(12.48gI·kg-1),while only the same volume of saline was infused intravenously in groups N.Five mice which were randomly selected in the two groups at10 min,20 min,30 min,40 min,50 min,1 h after drug treatment took blood and Collected renal tissue after anesthesia.ELISA to detct serum creatinine.To evaluate The renal tubules injury were examined by routine pathology and Semi-quantitative analysis of the degree of PTC injury by Immunohistochemistry of von Willebrand Factor.Ultrastructural changes of peritubule capillaries and renal tubules observed by transmission electron microscopy.Results:Compared with group N at the same time,there is no statistical difference in serum creatinine in group CM?32.33±10.78,36.15±5.34,32.29±10.12,37.02±12.38,32.58±10.14,33.35±11.11,33.59±9.29?,P>0.05.The positive expression of vWF in10min in group CM?0.15±0.01?was significantly higher than that in N group?0.10±0.01?,P<0.05.Compared with N group?0.84±0.20?,the score of renal tubule injury in 30min was significantly higher in group CM?5.66±0.61?,P<0.05.In CM group,the degree of renal tubule and PTC injury were significantly alleviated at 3060min?P<0.05?,and restored to normal in 72h.TEM:The damage of PTC endothelium and cell ultrastructure was prior to that of renal tubular epithelial cells,and both returned to normal within a certain period of time.Conclusion:PTC damage may be the initial link of renal injury induced by contrast media,its effect may be related to the dysfunction of cell barrier and energy metabolism.
