The testing of drugs plays an important role in ensuring the safety of drugs and improving the manufacturing process of drugs.Traditional drug testing generally uses conventional physical and chemical means to analyze the appearance,composition and physical and chemical properties of drugs,but it does not provide intuitive distribution of various components.Stimulated Raman scattering(SRS)microscopy,as a highly chemicalfree label-free microscopic tool,has been widely used in biomedical and life science fields in recent years,which has great potential in the detection and imaging of drugs.Here,we demonstrated high-speed and large-scale chemical imaging method based on spectral focusing hyperspectral SRS microscopy for identification and quantification of chemical contents in tablet.Firstly,we elucidated the technical principle of SRS microscopy by applying nonlinear coupled-wave equations.Moreover,we analyzed the noise of SRS signal,and developed method to reduce the noise background in SRS image.Finally,we built epi-SRS microscope,which achieved high-speed imaging of large samples.Secondly,we described the technical details of the spectral focusing based hyperspectral SRS microscope.In the optical system,SF57 glass rods were employed to induce enough dispersion for spectral imaging.We performed Raman shift calibration and intensity calibration to ensure the SRS spectra as identical to the original Raman spectra.Furthermore,we designed and built up-right SRS imaging system with epi detection,which detects the backscattering photons of pump beam.Finally,by using the spectral focusing hyperspectral SRS microscopy,we image the Sovaldi tablet with sub-micron resolution and large scale,and analyze the proportion and distribution of the main ingredients in tablets.The results showed great potential of SRS imaging technology for drug imaging and analysis.In summary,we developed high-speed and large-scale SRS microscope,which is promising for wide applications in field of drug study.
