Objective Malignant melanoma of the skin is a complex heterogeneous cancer with a polygenic etiology and is the leading cause of skin cancer-related deaths.More troubling is the steady increase in the incidence of the disease over the past decades.At present,surgical excision is still the only treatment method with early low-risk characteristics,but in the face of surgery with a high failure rate,the adjuvant therapy of vaccine is getting more and more attention.Cancer vaccines can reduce the growth rate of tumors and even induce apoptosis by activating body-specific immune responses.However,the immunogenicity of vaccines is low,so it is difficult to cause immune response of the body.Adjuvants are compounds or macromolecule complexes that can help regulate and enhance immune responses.They are combined with vaccines or cancer cells to improve their efficacy and increase antigen-specific responses.In addition,they enhance the effect of functional anti-tumor cells on the actual tumor site by facilitating the homing of effector lymphocytes.Radix scutellariae is a variety of biological activity of flavonoids,it has a variety of biological functions.Besides can remove oxygen free radicals in the body,still can improve blood circulation,lower cholesterol,such as efficacy.However,there is no report on its application as adjuvant.Therefore,in this study,we established a mouse melanoma model to detect the effect of baicalein on post-immune load.To determine whether baicalein can be used as an adjuvant for melanoma,the immune response of T cells in tumor mice was evaluated.Methods In the experiment,we used different concentrations of baicalein(50 mg/kg,100 mg/kg)and inactivated B16F10melanoma cell as experimental group,and with Freund's complete adjuvant and inactivated B16F10 as positive control.At the same time,the design of the different concentrations of radix scutellariae group(50mg/kg,100mg/kg)and separate inactivated B16F10 group,we are going to C57BL/6 mice were divided into 8 groups,each group of 10.Each group was immunized with B16F10 tumor cells subcutaneously after one week.Then we detected the tumor size and survival rate of mice bearing tumors,and the cytokines secreted by CD4~+T cells and CD8~+T cells from different groups of mice.Finally,CD8~+T cells were detected.To evaluate the inhibition of baicalein as adjuvant on melanoma and determine the concentration of baicalein.ResultsFlow cytometry analysis results showed that different concentrations of radix scutellariae element could up-regulate the expression of cytokines IL-2 and TNF-?secreted by CD4~+T cells,especially in 100 mg/kg baicalein and inactivated B16F10 tumor antigen group.Baicalein at different concentrations could up-regulate the expression of cytokines TNF-?and IFN-?in CD8~+T cells.Moreover,baicalein at 100 mg/kg and inactivated B16F-gamma were injected.The survival rate of mice in 10 groups was higher and the growth of tumors was slower.In the cellproliferation test,compared with the inactivated tumor cell group,the cell antigens of 100 mg/kg baicalein injection,inactivated B16F10 group and positive control group increased significantly.ConclusionBoth 50 mg/kg and 100 mg/kg baicalein can up-regulate the cytokines secreted by CD4~+T cells and CD8~+T cells to varying degrees.Among them,100 mg/kg baicalein has a better effect.Therefore,100 mg/kg baicalein has a strong effect on the activation of T cells in vivo,and 100 mg/kg baicalein as an adjuvant can increase the proliferation and killing activity of T cells.Both 50 mg/kg and 100mg/kg of baicalin as adjuvants can inhibit the growth of melanoma,among which 100 mg/kg of baicalin has better tumor inhibition effect,so 100 mg/kg of baicalin;100 mg/kg baicalein as an adjuvant can improve the survival rate of mice.Therefore,100 mg/kg baicalin may become a potential adjuvant for tumor vaccine.
