Objective To investigate the effect of down-regulation or increase of Mps1 expression on proliferation,apoptosis and metastasis of osteosarcoma cells in vitro.Methods Western blotting and qRT-PCR were used to detect the basic expression of Mps1,Mad1,Bub1 and CyclinB in osteosarcoma cells OS-732,U2 OS and osteoblast hFOB1.19.OS-732 with high expression of Mps1 osteosarcoma cells was divided into OS-732 inhibition group,si-Mps1 OS-732 group and Over-Mps1 OS-732 group.Western blotting and qRT-PCR were used to detect Mps1,Mad1,Bub1 and CyclinB.Factor expression differences,CCK-8 detected cell proliferation inhibition rate,flow cytometry to detect apoptosis rate,and scratch test to detect cell migration ability.Results The expressions of Mps1 and CyclinB in OS-732 and U2 OS were higher than those in hFOB1.19(P<0.05).The expression of Mad1 and Bub1 was lower than that of hFOB1.19(P<0.05),and the expression of Mps1 was the highest in OS-732.In the OS-732 inhibition group,the si-Mps1 OS-732 group decreased the expression of Mps1,CyclinB protein and mRNA(P<0.05),increased the expression of Mad1,Bub1 protein and mRNA(P<0.05),and the cell proliferation inhibition rate increased(P<0.05),the apoptosis rate increased(P<0.05),the cell migration ability decreased(P<0.05),and the si-Mps1 OS-732 group changed more significantly.The Over-Mps1 OS-732 group showed opposite results to the OS-732 inhibition group and the si-Mps1 OS-732 group(P < 0.05).Conclusions The osteosarcoma cell line OS-732,U2 OS has high expression of Mps1 and is positively correlated with the degree of malignant for osteosarcoma.The down-regulation of Mps1 expression can inhibit the proliferation and metastasis of osteosarcoma cells and promote apoptosis.The loss of Mps1 expression leads to the failure of SAC.The decrease of mitotic monitoring and false accumulation of osteosarcoma cells is the main mechanism of inhibition of OS-732 proliferation and metastasis and increase of apoptosis after targeting down-regulation of Mps1.
