Objective: To explore the analgesic effect by observing the effect of Fuyanshu Capsule on the writhing latency,writhing frequency and analgesia rate of acetic acid-induced pain mice model,and the mechanism by detecting the content of Prostaglandin E2(PGE2)?Nitric oxide(NO)in serum and Beta endorphins(?-EP)in brain tissue of the mice.Methods: 72 SPF Kunming mice were randomly divided into the low dose group of Fuyanshu,the middle dose of Fuyanshu,the high dose of Fuyanshu,the group of Kangfuyan capsules,the aspirin group and the blank group according to the random number table.And there were 12 mice in each group.The group began to gavage on the same day,once a day for 7 consecutive days.Sixty minutes after the end of the last gavage,each group was intraperitoneally injected with 0.9% glacial acetic acid to induce the writhing reaction of the mice.The writhing latency and the number of writhings were recorded within 20 min after the injection of the algogenic substance,and the analgesia rate was calculated.After observing the writhing behavior,8 mice were randomly selected from each group to collect the specimen.The serum levels of PGE2,NO and ?-EP in the brain were detected by ELISA.Results: 1.Compared with the blank group,the Fuyanshu high and middle dose groups can significantly prolong the writhing latency of mice and reduce the number of writhing in mice(P<0.05),and the writhing latency of Fuyanshu high dose group was significantly longer than that of the low-dose group(P<0.05),the lowest positive rate of writhing was 33.33%,the highest analgesia rate was 95.26%,andthere was no significant difference between the inhibitory effect of Fuyanshu Capsule on the writhing reaction of mice with that of the aspirin group or that of Kangfuyan group(P>0.05).2.Compared with the blank group,the high dose of Fuyanshu could significantly reduce the content of PGE2 in the serum of mice(P<0.05),and the effect of Fuyanshu Capsule on reducing PGE2 was not significantly different from that in the aspirin group or in the Kangfuyan group(P>0.05).3.Compared with the blank group,the content of NO in the serum of Fuyanshu high and middle dose group was significantly decreased(P<0.05).There was no significant difference between the effect of Fuyanshu Capsule on reducing NO with that of the aspirin group or that of the Kangfuyan group(P>0.05).4.There was no significant trend in the content of ?-EP in the brain tissue of each group compared with the blank group.There was no significant difference between the groups(P>0.05).Conclusions: 1.Fuyanshu Capsule can significantly inhibit the number of writhing in mice,shorten the latency of writhing,and has an obvious analgesic effect.2.Fuyanshu Capsule may exert an analgesic effect by lowering the level of PGE2.3.Fuyanshu Capsule may exert an analgesic effect by reducing the content of NO.
