| Objective To explore the effect of Fuzheng Huayu Jiangzhuo Tongluo Recipe on the expression of Smad7 and Smurf2 in kidney tissue of 5/6 nephrectomized rats,and to provide theoretical basis for Fuzheng Huayu Jiangzhuo Tongluo recipe in the treatment of renal fibrosis.Methods? Model establishment: SD male rats of clean grade were divided into operation group and sham operation group.The rats of sham operation group were fed adaptively for 7 days.The rats of sham operation group were treated with 5/6nephrectomy to establish the animal model of chronic renal fibrosis.They were randomly divided into model group,Chinese medicine intervention group(high,medium and low doses).The sham operation group separated the renal capsule and did not remove the renal tissue.? Drug intervention and sample treatment: 7 days after operation,rats in the intervention group of traditional Chinese medicine(high,medium and low doses)were fed with Fuzheng Huayu Jiangzhuo Tongluo recipe daily,rats in the model group were fed with distilled water daily,rats in the sham operation group were not treated with any treatment.The general condition,body weight,drinking water and diet of rats were observed.Rats were killed at 4,8 and 12 weeks after gastric lavage,respectively.Serum creatinine and urea nitrogen were detected.Index.? HE,Msson and PASM staining were used to observe the pathological changes of the kidney.The expression of Smad7 and Smurf2 in the kidney of rats was semi-quantitatively analyzed by immunohistochemistry.Results?The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on General Conditions of Experimental RatsBefore operation,rats in sham operation group,Chinese medicine intervention group(high,medium and low doses)and model group were all flexible,responsive,binocular,drinking water and diet were normal,body weight increased steadily,fur was bright,supple and white.Before the operation,there was no special abnormality in the general condition of rats in the sham-operated group;with the prolongation of time,the rats in the sham-operated group and the intervention group of traditional Chinese medicine(high,medium and low doses)were depressed in both eyes,gradually reduced in activity,curled up,drank less water and fed less,overreacted to the outside world,prone to irritation,rough yellowing hair,scratching marks on the tail,face and limbs,etc.In general,the wounds of rats in the intervention group(high,medium and low doses)were better than those in the model group.?The Effect of Fuzheng Huayu Jiangzhuo Tongluo Recipe on Serum Creatinine and Urea Nitrogen in Experimental RatsSerum creatinine: Compared with the sham-operated group,the serum creatinine in the model group increased significantly(P < 0.05).Compared with the sham-operated group,the serum creatinine in the Chinese medicine intervention group(high,medium and low doses)increased significantly(P < 0.05).There was a significant difference between the groups(P < 0.05).Compared with the model group,the rats in each Chinese medicine intervention group(high,medium and low doses)were significantly higher.The serum creatinine was significantly decreased,and there was significant difference between groups(P < 0.05).There was no significant difference among groups of Chinese medicine intervention(high,medium and low dose)rats(P > 0.05).Urea nitrogen: Compared with the sham-operated group,the urea nitrogen in the model group increased significantly(P < 0.05).Compared with the sham-operated group,the urea nitrogen in the model group increased significantly(P < 0.05).Compared with the sham-operated group,the urea nitrogen in the Chinese medicine intervention group(high,medium and low doses)increased significantly(P < 0.05).There was no significant difference between the pre-treatment group(high,medium and low doses)and the model group(P > 0.05).There was no significant difference between the groups(P > 0.05).There was no significant difference between the groups(high,medium and low doses)of Chinese medicine intervention group(P >0.05).? The Effect of Fuzheng Huayu Jiangzhuo Tongluo Recipe on Urinary protein in Experimental RatsCompared with the sham-operated group,the urinary protein in the model group increased significantly(P < 0.05).Compared with the sham-operated group,the urinary protein in the Chinese medicine intervention group(high,medium and low doses)increased significantly(P < 0.05).Compared with the sham-operated group,the urinary protein in the Chinese medicine intervention group(high,medium and low doses)increased significantly(P < 0.05).Compared with the model group,there was no significant difference in urinary protein between the low-dose group and the model group(P > 0.05).There was no significant difference in urinary protein between the groups of Chinese herbal medicine intervention group(high,medium and low dose)rats(P > 0.05).? The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on the Degree of Renal Fibrosis in Experimental RatsIn the sham-operated group,glomerular structure was intact,mesangium and basement membrane were not abnormal,tubular structure was normal,interstitial inflammation and fibrosis were not obvious;in the model group,glomerular mesangial cell proliferation,mesangial matrix increase,basement membrane thickening,renal capsule cavity enlargement and focal glomerular dura were observed in the rats of the intervention groups of Chinese herbal medicine(high,medium and low doses).Some renal tubular epithelial cells fell off,some renal tubules dilated,renal tubules atrophied,interstitial inflammatory infiltration and fibrosis were observed;the pathological changes of rats in each Chinese medicine intervention group(high,medium and low doses)were lighter than those in model group;there was no significant difference among the Chinese medicine intervention groups(high,medium and low doses).? The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on the Expression of Smad7 and Smurf2 in Experimental RatsSmad7: Sham-operated rats are widely expressed in kidney tissue.With the increase of time,the expression of rats in model group and Chinese herbal medicine intervention group(high,medium and low doses)is lower than that in sham-operated group(P < 0.05);the expression of rats in each Chinese herbal medicine intervention group(high,medium and low doses)is higher than that in model group(P < 0.05),and the difference is statistically significant(P < 0.05).There was no significant difference in Smad7 expression in renal tissue between the intervention group(high,medium and low dose)rats(P > 0.05).Smurf2: The expression of Smurf2 in kidney tissue of rats in sham-operated group was very little.With the increase of time,the expression of Smurf2 in kidney tissue of rats in model group and Chinese medicine intervention group(high,medium and low doses)was higher than that of rats in sham-operated group(P < 0.05).Compared with rats in model group,the expression of Smurf2 in kidney tissue of rats in each Chinese medicine intervention group(high,medium and low doses)was higher(P < 0.05).The expression of Smurf2 in kidney tissue of rats at different doses decreased significantly(P < 0.05).There was no significant difference in the expression of Smurf2 in kidney tissue among the Chinese herbal medicine intervention groups(high,medium and low doses),and there was no statistical significance(P > 0.05).? The correlation between the expression of Smad7 and Smurf2 and renal fibrosisThe correlation analysis showed that the expression of Smad7 was negatively correlated with serum creatinine and renal fibrosis,while the expression of Smurf2 was positively correlated with serum creatinine and renal fibrosis.conclusions?.Fuzheng Huayu Jiangzhuo Tongluo Recipe can delay the increase of creatinine and improve renal function in 5/6 nephrectomized rats,thus delaying the progress of renal fibrosis.?.Fuzheng Huayu Jiangzhuo Tongluo Recipe can reduce the degree of renal fibrosis in 5/6 nephrectomized rats,and its therapeutic mechanism may be through reducing the expression of Smurf2 and promoting the expression of Smad7. |
