Objective:With sympathetic nerve regulation as the starting point,the cardiac protective effect and mechanism of acupuncture neiguan point on microosmotic pump infusion of isoproterenol induced cardiac hypertrophy mice were investigated,providing a new method and theoretical basis for clinical treatment of cardiac hypertrophyexplore.Method:1.Grouping: 75 spf-grade male C57 BL mice were recruited,and the mice were divided into the blank control group,the myocardial hypertrophic model group,the acupuncture neiguan point treatment group,the acupuncture non-point treatment group,and the atenolol positive drug group according to the random number table method.2.Modeling: apart from the blank control group,isoproterenol(40mg/kg/d)perfusion microosmosis pump was placed under the back of mice,and the blank control group was treated with clean plastic of the same size as the microosmosis pump for sham surgery.3.Treatment:1.Acupuncture neiguan point treatment group: after inhalation anesthesia with isoflurane,0.17 mm × 7 mm stainless steel millineedle was used to directly Pierce bilateral neiguan points of mice with a depth of 2mm,and the needle was retained for 15 min.(2)acupuncture in the non-point treatment group: after isoflurane inhalation anesthesia,0.17 mm ×7 mm stainless steel filiform needle was used to acupuncture in the non-point distribution area of the tail root of the mice,and the acupuncture was kept for15 min.(3)atenolol positive group: after inhalation anesthesia for 15 min,atenolol(10mg/kg)aqueous solution was given,once a day.(4)myocardial hypertrophy model group and blank control group: isoflurane inhalation anesthesia for 15 min,once a day.Each group was treated continuously for 14 days.4.Determination: on the first and 15 th days of the experiment,the expression of norepinephrine(NE)in serum of mice in each group was detected by enzyme-linked immunosorption assay(Elisa)and compared between groups.On the 15 th day,the heart rate and echocardiography were used to detect the cardiac function of the mice in each group.The degree of myocardial fibrosis after HE staining and Masson staining was observed under ordinary optical microscope.The ultrastructure of cardiac muscle cells was observed by electrical microscope.Result:1.General status and death status: the blank control group was in good general status with regular breathing and no wheezing.In general,compared with the myocardial hypertrophy model group and the acupuncture non-point treatment group,the mice in the acupuncture neiguan point treatment group and atenolol positive drug group had better fur color and glossiness,and no rapid breathing.The mortality was lower in neiguan acupuncture group,but higher in model group,blank control group and atenolol positive group.2.Heart rate: compared with the blank control group,the heart rate of mice in the model group of myocardial hypertrophy and the non-acupuncture treatment group was significantly higher(P<0.05).Compared with the model group of myocardial hypertrophy,there was no significant difference between the treatment group of acupuncture neiguan point and the atenolol positive drug group,showing no statistical significance(P>0.05).3.Echocardiography: compared with the blank control group,the myocardial hypertrophy model group,acupuncture point treatment group after the left ventricular diastolic wall thickness(LVPWd),left ventricular posterior wall thickness of end systolic(LVPWs)were increased(P < 0.05),compared with myocardial hypertrophy model group,acupuncture in closing hole processing group of atenolol LVPWd and LVPWs value positive drug group were decreased(P < 0.05),the ejection fraction(EF)compared with no significant difference(P > 0.05).4.Observation by general optical microscope and transmission electron microscope: compensatory hypertrophy of myocardial tissue appeared in the model group of myocardial hypertrophy and the non-acupuncture treatment group compared with the control group,while the degree of myocardial cell and muscle fiber lesions in the treatment group of acupuncture neiguan point and atenolol positive drug group were significantly lower than those in the model group of myocardial hypertrophy.5.Serum norepinephrine(NE)levels in mice of each group: there was no difference in serum NE levels in mice of the five groups on the first day of the experiment(P>0.05).Within 15 days,compared with the blank control group,myocardial hypertrophy model group,acupuncture point treatment group NE expression level is higher,the difference is statistically significant(P < 0.05),compared with myocardial hypertrophy model group,acupuncture closed cavity in treatment group and the atenolol expression level is low,positive drug group of blood can be statistically significant difference(P < 0.05).Conclusion:1.Microosmotic pump perfusion of isoproterenol for 14 consecutive days can successfully produce a mouse model of myocardial hypertrophy.2.Acupuncture at neiguan point can significantly reverse the morphological changes of myocardial cells in mice with cardiac hypertrophy induced by microosmotic pump infusion of isoproterenol,and improve the degree of myocardial fibrosis.3.The reversal of myocardial fibrosis and cardiac protective effect of acupuncture on microosmotic pump infusion of isoproterenol induced cardiac hypertrophy in mice may be related to the inhibition of sympathetic nerve activity and the reduction of norepinephrine release.
