Objective:(1)The effect of Electroacupuncture(EA)on mechanical allodynia in rats model of lumbar disc herniation.(2)To observe the influence of EA on histomorphological changes of spinal nerve root in rats model of lumbar disc herniation.(3)To observe the influence of EA on JAK2-STAT3 pathway of spinal cord in rats model of lumbar disc herniation.Methods: Fourty-eight male SD rats were randomly divided into control group,sham-operation group,model group and EA group(each n=12).The model of Lumbar Disc Herniation was established by using autologous nucleus pulposus to the site close to the right L5 dorsal root ganglion.The mechanical withdrawal threshold was deceted by Von-Fair;histomorphological changes of spinal nerve root;Western blot were performed to explore the activation of JAK2-STAT3 pathway.EA group was applied at shenshu(BL23),dachangshu(BL25),huantiao(GB30),yanglingquan(GB34)and stat on day 3 after surgery,once a day and 20 min for 14 d.The mechanical pain threshold was measured.On day 9,16 after surgery,L4?L6 spinal cord was harvested.All the results were analyzed by statistical analysis.Results:(1)Compared with the control group and sham-operated group,the mechanical pain threshold of model and EA group were significant decreased(P<0.05);compared with the model group,the mechanical pain threshold of EA group was significantly increased.(2)The results of HE staining.control group:the nerve fibers is same size,arranged neat.sham group:the nerve fiber size is relatively consistent,nerve fibers arranged more neatly,no inflammatory cell infiltraton.model group: nerve fiber size isinconsistent,loose nerve fibers arranged,nerve fiber swelling,more common myelin segmental loss.EA group: nerve fiber tend ton normal relative size,relatively regular arrangement of nerve fibers,part of the nerve fibers swelling,showing a few myelin segments loss,a few nerve fibers naked.(3)compared with control and sham group,the content of p-JAK2?p-STAT3 in model group is increased;compared with model group,the content of p-JAK2?p-STAT3 in EA gourp is decreased significantly.Conclusion:(1)The model of lumbar disc herniation with autologous nucleus pulposus couldreduce the mechanical threshold,and EA could increased thethreshold of mechanical.(2)Transplantaton can cause lumbar 5 spinal nerve root fibers loosely arranged,demyelination,nerve fiber bare,EA "Shenshu","Dachangshu","Huantiao","Yanglingquan" can reduce the degree of spinal nerve root injury.(3)Autologous nucleus pulposus could lead to spinal cord p-JAK2,p-STAT3 protein content increased,EA can reduce p-JAK2,p-STAT3 protein expression,indicating that EA treatment of lumbar disc herniation rats may be through the regulation of the spinal cord JAK2-STAT3 signaling pathway.
