Objective: Triggering receptor expressed on myeloid cells 1(TREM-1)is a member of newly discovered innate immunity receptors,comprising an common domain of immunoglobulin superfamily.TREM-1 is found to express on neutrophils?monocytes and vascular endothelial as well.Previous evidence indicated that TREM-1 and its soluble form(Soluble triggering receptor expressed on myeloid cells 1,s TREM-1)is closely involved in autoimmune diseases and inflammatory reactions(both acute and chronic).But the relationship between TREM-1(s TREM-1)and coronary atherosclerosis is not clear so far.We aimed to investigate whether serum s TREM-1 is related to coronary artery disease(CAD)and to evaluate the biological effects of s TREM-1 and TREM-1 in cell experiments.Methods:1.This study included 263 consecutive patients with coronary angiographically diagnosed as CAD(CAD group),with 162 participants without CAD serving as controls(control group).Serum levels of s TREM-1 were determined in all participants.2.In cell experiments,the influence of s TREM-1 and TREM-1 on ox LDL-induced inflammatory reactions,monocyte-endothelial cell interaction and oxidative stress was evaluated in human umbilical vein endothelial cells(HUVECs).Results:1.Serum level of s TREM-1 was significantly lower in CAD patients than in controls(P<0.001).Moreover,s TREM-1 values were related to the number of diseased coronary arteries(Spearman's r =-0.413,P<0.001)and the severity represented by Gensini score(Pearson's r =-0.336,P<0.001).Multivariable logistic regression analysis revealed that decreased s TREM-1 was independent determinant of CAD(OR=0.608,P<0.001).2.In cell experiments,triggered TREM-1 synergized the effect of ox LDL-induced inflammatory reactions,monocyte-endothelial cell interaction and oxidative stress in HUVECs,which can be partly inhibited by recombinant s TREM-1 protein.Conclusions:1.This study demonstrates that decreased serum s TREM-1 level is significantly associated with the presence and severity of CAD.2.Triggered TREM-1 synergizes the effect of ox LDL-induced inflammatory reactions ?monocyte-endothelial cell interaction and oxidative stress in HUVECs.s TREM-1 restrains inflammatory reaction in endothelial cells,suggesting that it might be a potential vascular protective factor.
