ObjectiveTo explore the relationship between the expression of bone morphogenetic protein-9(BMP-9)and the prognosis of breast cancer patients and other clinical data,and screen out long-chain non-coding RNA related to BMP-9 expression,so as to provide experimental basis for exploring the mechanism of BMP-9 affecting the prognosis of breast cancer patients.MethodsThe clinical data of breast cancer patients were analyzed by Kaplan M and BC-GenE database.The changes of lncRNA expression profiles in MDA-MB-231/BMP9 cells and control group(MDA-MB-231/GFP)were detected by microarray technology.The selected lncRNA was verified by real-time fluorescence quantitative PCR(q-PCR)in MDA-MB-231/BMP9 cells and MDA-MB-231/GFP cells.The functions and pathways of lncRNA with altered expression levels were analyzed by DAVID database.ResultsThere was significant difference in relapse-free survival between BMP9 high expression group and low expression group(P < 0.05);BMP9expression was related to age and molecular subtypes of breast cancer patients(P < 0.05),but not to receptor type,lymph node status,SBR histological grade,basal-like/triple negative breast cancer(P > 0.05).The expression profile of lncRNA in MDA-MB-231/BMP9 cells changed significantly compared with the control group.Q-PCR showed that the expression levels of LINC00461,IQCA1,LOC440173,ANKRD36BP2 and LHFPL were down-regulated(P < 0.05),while those of LINC00638,LINC00486 and LINC00443 were up-regulated(P < 0.05).Differentially expressed lncRNA participates in the formation of cell components such as membrane and skeleton,or acts as signal transduction molecules between cells.ConclusionThe prognosis of breast cancer patients with high BMP9 expression is better than those with low BMP9 expression,and the expression of BMP9 is related to age and molecular subtypes.The expression level of LncRNA in MDA-MB-231/BMP9 group was significantly different from that in control group,suggesting that lncRNA may play a role in the prognosis of breast cancer patients affected by BMP-9.
