Objective : To evaluate the efficacy and safety of S-1 versus capecitabine in advanced breast cancer by meta-analysis.Methods: The database of Pubmed,The Cochrane Library,EMbase,CKNI,Wangfang were searched by computer.According to the inclusion and exclusion criteria the documents and the data were selected, the meta-analysis was performed by RevMan 5.3 software. Results: 12 RCTs involving 829 patients were identified.Meta-analysis showed that S-1 compared capecitabine can improve the disease control rate of advanced breast cancer[OR= 1.64,95%CI(1.12,2.40),(49)=0.01],the difference was statistically significant. But there was no significant difference in objective remission rate[OR = 1.34,95%CI(0.99,1.81),(49)=0.05].In terms of safety, the incidence of malignant vomiting[OR= 0.32,95%CI(0.16,0.65),(49)=0.002],hand-foot syndrome[OR= 0.14,95%CI(0.06,0.30),(49)<0.00001],S-1 was significantly lower than capecitabine, the results were statistically significant, and there was no significant difference in leukopenia[OR= 0.72,95%CI(0.38,1.38),(49)=0.32].Conclusion: S-1 can effectively improve the disease control rate, and has better safety in malignant vomiting and hand-foot syndrome.
